Objective: To study the epidemic situation and characteristic of K14.1 Alleles of Human herpesvirus 8 in Kaposi′s Sarcoma in Xinjiang, and Primary discuss correlations between different clinics of Kaposi's sarcoma and different subtypes. Methods: HHV-8 DNA was extracteded with phenol-chloroform-isoamyl alcohol from 32 cases formalin-fixed , paraffin-embedded tissue from the People′s Hospital of Xinjiang Uygur Autonomous Region . The K14.1 gene of HHV-8 DNA were amplified by nested-PCR. Sequentially,the K14.1 allele was confirmed . Correlations between different clinics of Kaposi's sarcoma and different subtypes were determined by Fisher's exact probabilities.Results: HHV8 DNA was detected in 27 (84.38%) of 32 cases . HHV-8 DNA was detected in 5 cases AIDS-KS (100%). PCR-based studies of the K14.1 genomic region indicate that twenty-four of these strains were of the P subtype while three belonged to the M subtype. Five cases AIDS-KS patients were respectively infected by the P allele. But the three M allele strains were extracteded from classical Kaposi′s Sarcoma patients. The analysis of the biologic and clinical data regarding evolution of KS in our patients show a trend toward no differences between patients with HHV-8 subtype P<0.05) .The discrepancy of K14.1 alleles of HHV-8 among different clinicoepidemiologic forms of Kaposi′s Sarcoma was not significant (P >0.05). Conclusion: 84.38% of KS patients were infected by HHV-8 P allele of HHV-8. HHV-8 infection plays an important role in the pathogenesis of KS in Xin Jiang . But there were some patients infected by M allele .Remarkably, K14.1 M allele for the first time at home . AIDS-KS patients in Xin Jiang tend to younglings. The study be uncertain of correlations between different subtypes and clinicoepidemiologic forms of KS in Xin Jiang . Therefore,it is very necessary to collect added AIDS-KS samples to study .
