Objective: To study the epidemic situation and characteristic of k12/T0.7subtype ofHuman herpesvirus8in Kaposi’s Sarcoma in Xinjiang, and primary discuss correlationsbetween different clinics of Kaposi’s sarcoma and different subtypes.Methods: HHV-8DNA was extracteded with phenol-chloroform-isoamyl alcohol from30cases formalin-fixed,paraffin-embedded tissue from the People’s Hospital of XinjiangUygur Autonomous Region. The k12/T0.7gene of HHV-8DNA were amplified bynested-PCR, and then were bi-directional sequenced. Deduced amino acid sequences withthe software DNAMAN were aligned manually with the program Clustal W. The phylogeneticanalysis were carried out by using the software PHYLIP package,and then the k12/T0.7subtype was confirmed. Correlations between different clinics of Kaposi’s sarcoma anddifferent subtypes and single nucleotide polymorphism were determined by Fisher’s exactprobabilities.Results:(1)HHV-8DNA was detected in22(73.33%) out of30cases in Xin Jiang.HHV-8DNA was detected in7cases AIDS-KS (100%).(2)Phylogenetic studies, based onthe partial k12/T0.7gene nucleotide sequence analysis, indicate that ten strains belong to theA subtype and twelve belonging to the A/C group.(3)The nucleotide sequence of k12/T0.7(407bp)differs by0to0.35%between HHV-8isolates. The A and A/C subtype differ fromeach other by0.16%to0.35%.(4)The different k12/T0.7subtypes of HHV-8among mucosadamage of Kaposi’s Sarcoma were not significant (P>0.05). The subtype A and A/Cdisplayed frequent mucosal are29.41%and31.25%.(5)The different k12/T0.7subtypes ofHHV-8between classical Kaposi’s Sarcoma and AIDS Kaposi’s Sarcoma were notsignificant (P>0.05).(6) HHV-8k12/T0.7were not correlated with increased susceptibility tomucosa damage and clinical classification of Kaposi’s Sarcoma(P>0.05).Conclusion:(1)The majority of Kaposi’s Sarcoma patients in Xin Jiang were infectedby the A and A/C subtype of HHV-8.(2)The different k12/T0.7subtypes of HHV-8havenothing to do with Clinical stage、clinical classification and mucosa damage of Kaposi’sSarcoma.
