Objective:To understand the situation of prevalent and regional distribution of KSHV innormal population in Anhui Province, we conducted the research on the infection ofKSHV and preliminary analysis of risk factors for KSHV infection in Anhui Province.The work will help to provide a scientific basis for the continuous survey of the KSHVepidemiological in China and analysis of related molecular biology of KSHV, and thuspreventing the occurrence of KSHV and cutting off the route of transmission.Methods:The KSHV genes of ORF65, ORF73and ORF K8.1were constructed inprokaryotic expression vector pQE-80L. The fusion proteins were expressed in E.Colistrain BL21(DE3) and purified by Ni2+affinity chromatography. The expressedprotein and its purified product were identified by sodium dodecylsulfate–polyacrylamide gel electrophoresis (SDS-PAGE). Using these purifiedrecombinant proteins, the serologic diagnostic ELISA method was established andoptimized. The epidemiology of KSHV in1008subjects in Bengbu,1000subjects inHefei,1006subjects in Anqing in general population and158subjects with HIV-1infection in some places in Anhui were investigated based on the established ELISA.The results were analyzed by SPSS software.Result:①The seroprevalence of KSHV in the general population in Bengbu was4.3%andslightly higher in female than male subjects (4.6%vs.3.9%). Statistical analysis showed that seroprevalence of KSHV has no differences in the gender and the age.Subtype analysis showed two KSHV K1sequence in Bengbu, Anhui Province had aclose relationship with the K1sequence of EMA7, and these two subtypes belong tosubtype A.②The seroprevalence of KSHV in the general population in Hefei was4%, themale-positive rate was3.9%and female-positive rate was4.6%. Statistical analysisshowed that seroprevalence of KSHV has no differences in the gender and the age, butKSHV infection and tumor occurrence had a significant correlation (P=0.007).③The seroprevalence of KSHV in the general population in Anqing was5.3%, themale-positive rate was5.3%and female-positive rate was5.6%. Statistical analysisshowed that KSHV infection and syphilis infection had a significant correlation (P=0.03), but it had no differences in age, gender, blood type and hepatitis B infection.④The seroprevalence of KSHV in HIV-positive in gay men was12%in some placesin Anhui. Statistical analysis showed that KSHV infection and sexually transmitteddiseases infection had a significant correlation (P=0.002).Conclusion:These results indicated that the infection rates of KSHV were low in the generalpopulation in Anhui Province. And the seroprevalence of KSHV had no differences inthe gender and the age. The sample analysis showed that the infection of KSHV had asignificant correlation with tumor and syphilis infection. Thus cancer and syphilisinfection were the factors which affected KSHV infection rates. KSHV antibodypositive rate of HIV-positive gay men was higher than the general population. HIVinfection was one of the risk factors for KSHV serum antibody positive. And thesesamples of KSHV positive rate and sexual history had a significantly correlation,indicating that sexually transmitted diseases was one of the risk factors for KSHV and sexual transmission in KSHV transmission is an important way.
