Part I: Effects of vitamin D deficiency during pregnancy on femoral VDR expression of offspringObjective: To study the expression of VDR on the femurs of offsprings whose mothers are deficient of vitamin D.Methods: 36 adult female SD rats were chosen and allocated randomly into control group (18 rats) and model group (18 rats). The control group received vitamin D fodder with normal solarization and the model group received non-vitamin D fodder without solarization for 2 weeks. Then the female rats in two groups copulated with the male normal rats. The left femurs were collected from the 20-day embryos and the 1-day and 7-day newborns. VDR expression was measured by FQ-PCR and Western blotting.Results: The VDR expression of the model group is lower than the control group in E20d,B1d and B7d, respectively (P<0.05). Conclusion: The lower expression of VDR on the femurs of offsprings is due to vitamin D deficiency during the pregnancy, which may be a reason of offsprings'skeletal deformity. Methods: 36 adult female SD rats were chosen and allocated randomly into control group (18 rats) and model group (18 rats). The control group received vitamin D fodder with normal solarization and the model group received non-vitamin D fodder without solarization for 2 weeks. The expression of Bone Development-related Genes BMP-2,Cbfa1,Dmp1 was measured by FQ-PCR.Results: The expression of BMP-2 mRNA,Cbfa1 mRNA and Dmp1 mRNA in the model group is lower than the control group in E20d,B1d and B7d, respectively (P<0.05).Conclusion: The lower expression of bone development-related genes BMP-2,Cbfa1,Dmp1 on the femurs of offsprings is due to vitamin D deficiency during the pregnancy , which would affect the offsprings'bone development.
