[Objective]①In vitro establishment of human periodontal ligament fibroblast cells(HPLFs) model and in order to study the role of cytokines in HPLFs in regulating cell functions and providing experimental conditions.②Purpose of this study is to observe the gene expression of the hepatocyte growth factor (HGF) in HPLFs by transforming growth factor- beta 1 (TGF-β1) and interleukin-1β(IL-1β),and provide new document to the research of the effect of HGF network to HPLFs.[Methods]①We recoveried, isolated and cultured HPLFs from the cryopreserved third generation in vitro, observed the biological characteristics of HPLFs and the stable HPLFs model will be constructed in vitro.②Choose the fifth generation of good shape,vigorous growth of HPLFs,and randomly divided into experimental groups and control group,the experimental groups were stimulated with IL-1βof 0.1ng/ml,1ng/ml,10 ng/ml,50 ng/ml,100 ng/ml on cells respectively,the control group without IL-1βintervention replaced with fresh DMEM medium. After 48h, using RT-PCR detected the HGF gene expression of HPLFs, and optimized the best stimulation concentration of IL-1β.③Choose the fifth generation of good shape,vigorous growth of HPLFs,and randomly divided into experimental groups and control group,the experimental groups were joint stimulated with IL-1βof 10ng/ml and TGF-β1 of 1ng/ml,5ng/ml,10ng/ml,50ng/ml,100ng/ml on cells respectively, the control group with IL-1βof 10ng/ml . After 48h, using RT-PCR detected the HGF gene expression of HPLFs.④Using medical image analysis system analyzed the results of image and data collection, data results were expressing of x ? s, using SPSS13.0 software analyzed, groups were compared using analysis of variance, multiple comparisons with LSD-t test , P <0.05 was considered statistically significant.[Result]①The IL-1βalone, the HGF content in the supernatant of HPLFs more than in the blank control group, and found the best markedly concentration of IL-1βis 10ng/ml(P<0.05).②When a certain concentration of TGF-β1 and 10ng/ml IL-1βwere joint stimulated HPLFs, the HGF gene expression in HPLFs than separate the role of IL-1βwere lower(P<0.05).[Conclusion]①The role of IL-1βsignificantly induced the gene expression of HGF in HPLFs;②When IL-1βof 0.1,1,10 ng / ml, the gene expression levels of HGF with IL-1βconcentration increased in a dose dependent manner; and compared with IL-1βof 10ng/ml, when IL-1βof 50,100ng/ml, the gene expression of HGF was no significantly increased.③The role of IL-1βsignificantly induced the gene expression of HGF in HPLFs, When joint stimulation of IL-1β and TGF-β1, the gene expression levels of HGF were reduced with the increasing concentrations of TGF-β1. TGF-β1 can inhibite the IL-1βinduced the gene expression of HGF in HPLFs.
