Objective: To establish a determination method for Senegenin and Polygalacic acid in Radix Polygalae and its preperations by capillary electrophoresis; To establish the high performance capillary electrophoresis (HPCE) fingerprint chromatogram of genuine Radix Polygalae from Shanxi and get the control chromatogram, then to compared the similarity of fingerprint chromatogram of Radix Polygalae from different place.Methods: The determination method for Senegenin and Polygalacic acid in Radix Polygalae and its preperations by capillary electrophoresis: the separation was carried out in an uncoated fused silica capillary (75μm×57 cm). The running voltage was 25 kV. The buffer solution was 20 mmol/L Na2B4O7 containing 20% methanol, and the wavelength was 210 nm. The method of high performance capillary electrophoresis (HPCE) fingerprint chromatogram of genuine Radix Polygalae: the separation was carried out in an uncoated fused silica capillary (75μm×57 cm). The running voltage was 18 kV. The buffer solution was 20 mmol/L Na2B4O7 containing 25% methanol(v/v), and the wavelength was 210 nm. Data and chromatogram were deal with"Computer Aided Similarity Evaluation"software in order to analyse the similarity of fingerprint chromatogram of Radix Polygalae from different places.Results: The method was validated over linear ranges of 13.25~212μg/mL ( r = 0.9971) for senegenin and 12.5~200μg/mL ( r = 0.9977) for polygalacic acid. The average recoveries of senegenin and polygalacic acid in Radix Polygalae were 101.5% and 101.4%. The average recoveries of senegenin and polygalacic acid in Naolibao wan were 100.5% and 100.6%. The average recoveries of senegenin and polygalacic acid in Guipi wan were 101.5% and 100.4%. The average recoveries of senegenin and polygalacic acid in Tianwang buxin wan were 100.5% and 100.3%. The average recoveries of senegenin and polygalacic acid in Bushen yinao capsule were 100.2% and 100.2%. The average recoveries of senegenin and polygalacic acid in Baizi yangxin wan were 100.2% and 100.3%.The control fingerprint of HPCE of Radix Polygalae was established, and the similarity of fingerprint chromatogram of Radix Polygalae from different places were compared.Conclusion: This method of determination for Senegenin and Polygalacic acid in Radix Polygalae and its preperations by capillary electrophoresis is proved to be simple, rapid and accurate, and can be used as a new method for the quality control of Radix Polygalae. The method of high performance capillary electrophoresis (HPCE) fingerprint chromatogram of genuine Radix Polygalae is accurate, and can serve as a effective method for the quality control of Radix Polygalae.
