Objective:to study effect of ovary-protecting decoction on E2,P, LH levels of rats with controlled superovulation, as well as its effect in different dosage, thus offer theoretical evidence for TCM adjunctive therapy of IVF-ET.Method:all experimental rats were randomized into 5 groups, modeling group, low-dose medicinal group, moderate-dose medicinal group, high-dose medicinal group and normal group. The former four groups were given intraperitoneal injection of Alarelin, and simultaneously all medicinal groups were given intragastric administration of low, moderate and high dose Chinese medicinals respectively. Till the 9th day, HMG was injected to the former four groups, HCG was injected after 48 hours, and all rats were mated into cages at ratio of 2:1 of female to male. The rats which were discovered with vaginal plugs the next day was marked as first-day pregnancy. Then all rats were executed by neck break in batches, at 48 hours after HCG injection and on the fifth day of pregnancy, venous blood were withdrawn. For normal group, the rats were given both intragastric administration and intraperitoneal injection of saline, and were executed in batches 48 hours after ovulation and on fifth day of pregnancy, venous blood were withdrawn as well.Results:1. On the 5th day of pregnancy after superovulation, E2 levels were elevated, its level of modeling group and all medicinal groups was higher than that of normal group in significant difference (P< 0.05), ovary-protecting decoction can raise E2 level, however compared with modeling group, there was no significant difference (P> 0.05). No significance was found (P> 0.05) among low, moderate and high-dose groups; 2. On the 5th day of pregnancy after superovulation, P levels were elevated, with modeling group higher than normal group, the difference was significant (P<0.05), ovary-protecting decoction can raise P levels, which of all medicinal groups were higher than normal group with significant difference (P< 0.01). Among them, high-dose and moderate-dose group had higher levels of P as compared to modeling group, their differences were significant (P< 0.05), whereas the difference between low-dose group and modeling group was not significant (P> 0.05). P levels of high-dose and moderate-dose group were higher than low-dose group, the difference was significant (P< 0.05); 3. ovary-protecting decoction can also increase LH levels of 5-day pregnancy rats after superovulation, significant difference was found between all medicinal groups and normal group (P< 0.05). Level of modeling group was higher than normal group in significant difference (P< 0.05), levels of all medicinal group were higher than modeling group in significant difference (P< 0.05), and levels of high and moderate-dose groups were higher than low-dose group, the difference were significant (P< 0.05) while no significant difference was found between levels of high-dose group and moderate-dose group (P> 0.05). 4. E2 levels of all groups at 5th day of pregnancy (implantation window phase) were higher than values 48 hours after HCG injection. There were significant difference among all groups (P<0.05); 5. P levels of all groups at 5th day of pregnancy (implantation window phase) were higher than values 48 hours after HCG injection. There were significant difference among all groups (P< 0.05); 6. LH levels of all groups at 5th day of pregnancy (implantation window phase) were higher than values 48 hours after HCG injection. There was significant difference among all groups (P< 0.05).Conclusion:1. ovary-protecting decoction can increase E2, P, LH levels, especially P, LH values, in order that superovulated follicles will become more mature, making it more favorable for embryo implantation; 2. Through global regulation, ovary-protecting decoction make P/E2 much closer to its biological level which is beneficial for pregnancy rate; 3. Optimization of E2, P, LH levels by ovary-protecting decoction has certain dose-effect relationship with medicinal concentration.
