Experimental Study On The Impact Of Allergic Contact Dermatitis. Shufeng Drink

Purpose:To study the pharmacology effect and the part of immunology mechanism of Allergic Contact Dermatitis(ACD) intervened by Shufeng decoction, the model of ACD mice was established. In this research, the auricular swelling degree,weight difference,cell factors and pathology exchanges,etc of mice were mesured.It can provide rationales on the clinical application of Shufeng decoction.Material and method:50 mice were randomly divided into 5 groups: normal saline group, ACD model group, Shufeng decoction group ,loratadine group, mixed group of TCM and western medcine.The mouse model of allergic contact dermatitis was formed by conventional methods,each mouse irrigated stomach 0.4ml every day, Executed mice after nine days.Through a comparison of the auricular swelling degree and weight difference of the mice in each group,there was a preliminary evaluation for the role of Shufeng decoction; To observe the effects of Shufeng decoction on cell factors, enzyme-linked immunosorbent assay was performed to examine the levels of tumor necrosis factor-α(TNF-α) in mice of each group,immunohistochemistry technique was used to detect the expressions of TNF-αand interleukin 12(IL-12) in mice of each group.HE pathology examination was used to determine the effects of Shufeng decoction on the tissue pathology exchanges of ACD mice.Through index difference comparison of every group mice, the therapy effects and mechanisms of Shufeng decoction to ACD were evaluated.Results:1.Shufeng decoction reduced the auricular thickness difference and swelling degree,compared with the ACD group,the difference was statistically significant(P<0.05).2.Shufeng decoction group ,Loratadine group, mixed group of TCM and western medicine all declined the levels of TNF-αin the serum of the ACD mice, compared with the ACD group,the difference was statistically significant(P<0.05); Shufeng decoction group ,Loratadine group, mixed group of TCM and western medcine all reduced the expressions of TNF-αand IL-12 in the tissue of the ACD mice, compared with the ACD group,the difference was statistically significant (P<0.05);The results showed that no significant difference on levels of TNF-αin the serum and the expressions of TNF-αand IL-12 in the tissue was found between Shufeng decoction group and Loratadine group.3.Pathology sections of skin tissues from ACD model group displayed that large quantities of inflamation cells infiltrated in the epidermis and basal lamina and interstitial edema was existed. By the intervention of Shufeng decoction, the inflamation cells infiltrated in the epidermis and dermis were obviously alleviated and interstitial edema was declined markedly.Conclusion:1.Shufeng decoction can obviously reduce the auricular thickness difference and swelling degree of ACD mice and inhibit the inflammation reaction in the auricular sites of ACD group.2.Shufeng decoction declined the levels of TNF-αin the serum of the ACD mice, reduced the expressions of TNF-αand IL-12 in the diseased tissue of the ACD mice.3.Shufeng decoction alleviated inflamation cells infiltrated in the auricular tissues of ACD group.4.This study shows that Shufeng decoction has certain effect to ACD, its mechanism may be related with which Shufeng decoction can regulate some cytokines secretion, which regulates Th1/Th2 imbalance.
