Study On The Effect Of Interferon Induced Protein 10 On Cytokines In The Model Of Allergic Contact Dermatitis

Objective:1.To study the function of Chemokines IP-10 in ACD through observing their expression in ACD.2.To discuss the molecular mechanism of IP-10 in the development of Allergic Contact Dermatitis through observing the effect of IP-10 on expression of IFN-γ,IL-4 mRNA in vitro culture lymphocyte of ACD mouse.Method:Establish the ACD mouse model with dinitrochlorobenzol(DNCB),and carries on the following experiments:1.Detect the expression of IP-10 mRNA in lymphocyte of ACD mouse by RT-PCR.2.Detect the expression of IFN-γ,IL-4 in vitro culture lymphocyte of ACD mouse which incubation 48h with different density IP-10(0,10,20,100 ng/mL) by RT-PCR.Results:1,The expression of IP-10 mRNA in lymphocyte of ACD mouse is obviously higher than that of the control group.(P < 0.01);2,Compared with the control group,20 and 100 ng/mL concentration groups of IP-10 could remarkably promote the expression of IFN-γin vitro culture CD4~+T cell of ACD mouse(P<0.01和P<0.05),while there was no significant difference between the 10 ng/mL concentration group of IP-10 and control group.There was significant difference among three different concentration groups of IP-10 in their promoting the expression of IFN-γin vitro culture CD4~+T cell(P<0.01和P <0.05).3.There was no significant difference among the three different concentration groups of IP-10 on the expression of IL-4 in vitro culture CD4~+T cell,as well as when they compared with the control group(P>0.05).Conclusion:1.IP-10 highly express in ACD,which confirmed that IP-10 participate the development of ACD. 2.IP-10 may obviously to promote the expression of IFN-γmRNA.3.IP-10 was no obvious change of IL-4 mRNA expression in ACD.4.IP-10 may play an important role in the imbalance between Th1 and Th2,which profit Th1 cell recruit to the spot where antigen stimulated,and then Th1 inflammation response occur;which promotes the development of ACD.
