Objectives: The purpose of this study was to clarify whether high hydrostatic pressure affects the nitric oxide synthase (NOS) acivity and P-selectin secretion of cultured human umbilical vein endothelial cells (HUVECs), and to observe the effects of Captopril (Cap) and L-Arginine(L-Arg). Methods: The intima of human umbilical vein was digested by 0. 1% collagenase I. The 4th~6th passages of HUVECs were cultured in a high-tension apparatus filled with 5%co2and 95%air atatmosphere (0mmHg), 120 or ISOmmHg pressure Tor 24 hours. Under each pressure, one group was treated with Cap(10~5mol/l), one with L-Arg(10~5 mol/1), another as control. The NOS activity was measured with ELISA and the percentage of P-selectin positive HUVECs was evaluated by flowcytometric analysis. Results: Compared with 0 mmHg condition, the NOS activities were obviously lower (1.600 ?0.232U/ml and 1.109 ?0. 251U/ml vs 2. 316 + 0. 185U/ml, both RQ. 01) and the percentages of P-selectin positive HUVECs were higher (1.54 + 0.23% and 2.82?. 72% vs 1.24 + 0.16%, KO. 05 and KO. 01) in 120mmHg and ISOmmHg pressure conditions. In 0 mmHg condition, Captopril and L-Arg did not influence the NOS activities and the percentages of P-selectin positive HUVEGs, but in 120 and 180 mmHg conditions, the NOS activities in L-Arg groups (2. 026 + 0. 184U/ml vs 1. 600 + 0. 232U/ml, and 1. 700 + 0. 260U/ml vs 1.109 ?. 251U/ml, both KO. 01) and in Cap groups (1. 977 + 0. 226 U/ml vs 1. 600 + 0. 232U/ml, and 1. 445 + 0. 270U/ml vs 1.109 + 0.251 U/ml, both RO. 05) were higher than those of the control groups respectively; The percentages of P-selectin positive HUVECs were obviously lower in L~Arg groups (1.27 + 0. 15% vs 1.54 + 0. 23% and 1.89 + 0.69% vs 2.82 + 0.72 %, both KO. 01) and in Cap groups (1. 30 + 0. 14% vs 1. 54 + 0. 23%, KO. 05 and 1.72 + 0.38% vs 2.82 + 0.72%, K0.01). Conclusions: High hydrostatic pressure promotes.HUVECs P-selectin secretion and decreases the NOS activity. The changes are more obvious in ISOmmHg group than those of the 120 mmHg group. The treatment of Cap and L-Arg can attenuate the abnormal reactions induced by high hydrostatic pressure.
