Objective: Previous wik has suggested that many Chinese medicines have significant effect in cardiovascular diseases, they could relieve the damages of vanous ettological factor on vascular endothelial cells. In this study, we observed the inlluonces of function of Human tuiibilical vein endolhelial cells cultured with different press and hydroxyi ilee mdicalsQ-IFR) conditions in vitro, and investigated the efl~cts of intervention of Puemrin and Captopril on HUVECs cu~d ~itb the same conditions. 2 Melliods HUVECs were isolated by Jaffe抯 way and the endoftielial cells of 4tIm髏h passages was used The e.~per駈ont was divided into pressure and HRF groups. HUVECs were e~osed to atmosphere (OmmHg, 370C~ 50/vCO2),middle pressure (l2OmmHg 37X2, 5~C02) ,high pressure (lSOmmHg, 370(2,50/0(202) and FIRF(OFt: 10~rnoliL, 370(2, 5%CO2)respectively and maintained in RPMI- 1640 supplemented ~ith 10% calf sennn and subsUates for upto 48 horns after intervened by dmgs, then the ca2lenis of NO and the level of activity of ACF in medium were detected 3 Results: The middle pressure could increase NO and ACE excrctxm of VECs compared ~tb ihe ncumai pressi~ The high pressure and HRF could reduce NO and increase excretion of VECs respectively compared ~th nonnal pressure. Puerann could increase NO and reduce ACE excrelion of VECs compared with control group under the condition of high pressure and HRF Captopril could increase NO and reduce ACE excretion of VECs compared wilh control group nmder the high pressure 3 4 Conclusions: (1) Harvest and maintenance of HUVECs w~ peifoimed by modified Jaffe抯 ctdttn~ mediods. (2) The contents of NO and die activity of ACE excreted by VECs w~ different under different pressiie and FIRE condition, The high pressure and 1-IRF could reduce NO and increase ACE excretion of VECs. (3) Puerarfri could relieve die damage of high pressin~e and HRF on VECs. (4) Captopiil could relieve the damage of high pressure on VECs.
