Objective: To investigate the effect of retinoic acid modified intr aocular lens (RAM IOL) implantation on the proliferation, collag en synthesis of rabbit residual epithelial cells (RLECs) and its fe asibility. Method:1 .polymethylmethacrylate intraocular lenses(PM MA IOLs)were surface-modified with 5 i~ g or 10 1-t g all-trans ret inoic acid(atRA); 2. 26 New Zealand white rabbits were divided into normal group, control group, 5 i-i g RAM JOL implanted gr oup and 10 i~ g RAM IOL implanted group, receiving no therap y, implantation of no modified PMMA IOLs, 5 ~t g RAM lOLs a nd 10i~tg RAM IOLs on both eyes. At 2, 7, 15 and 30 days p ostoperatively, atRA concentration in aqueous humor was detecte d. The effects of RAM IOL implantation on the expression of p roliferating cells nuclear antigen (PCNA) and type V collagen of RLECs were examined by SABC immunohistochemical method; 3.Histopathological examination on local ocular tissues were per formed to evalued the toxic and side effects of atRA. Result: 1. The mean atRA concentration in aqueous humor of 4 trial group s was 3.10 ?0.l6ng.rnF?3.46 ?1.04 ng.mF?,74.18 ?3.27 ng.mli?a nd 106.64 ?2.08 ng.mi?respectively, and the atRA concentration of 5 it g RAM JOL implanted group, 10 1-?g RAM IOL implante d group maintained stably in 7 days and 15 days postoperatively; 2 2.Compared with control group, the expression of PCNA and typ e V collagen were lower in 5 1.t g RAM JOL implanted group an d 10 it g RAM JOL implanted group, and there were significant difference among them(p<0.0 1); 3 .However, the difference of da mage to corneal endothelial cells among them had no statistical significance(p>0.05), and the structure of iris, ciliary body, retina were intact, without obvious toxic reaction. Conclusion: RAM I OLs could maintain high atRA concentration in aqueous humor f or a long period after implantation of intraocular lens and had e ffective inhibition on proliferation and collagen synthesis of resid ual lens epithelial cells but little toxity. So it may become an ef fective method to prevent posterior capsule opacification for clini cal application.
