Puerarin Identification And Quality Evaluation Method Of Compound Preparation

Objective: To establish a complete qualitative and quantitative analysis system of Radix Puerariae, and screen out a rational extraction process for the main effective components in Radix Puerariae. Methods: The uniform design was used to study the effects of variety in extracting and selected the perfect parameters, with the extraction efficiency of Puerarin and flavonoids as index.. The Puerarin content of Radix Puerariae was determined with TLC-scanning and RP-HPLC. Results The optimum process was chosen by uniform design .The optimum water-extracting condition was as follows: with 12 times water in volume decocting 3 times, once every two hours . The optimum alcohol extracting condition was as follows: 12 times amount of 60% alcohol solution, soaking for 2.5 hours, refluxing and extracting 3 times, once every 2 hours. TLC and HPLC: by uniform design, chromatographic analysis elevated the degree of separation and repeatability. Conclusions: The optimized extraction procession is feasible and rational .The contents testing methods have highly accuracy and repeatability. Thy can be used effecting in quality control of Radix Puerariae in compound preparations.
