Cordyceps sinensis is among the most valued medicial fungi in traditional Chinese medicine It's used to strengthen and rebuild physical strength, nourishing nerve, vigorate energy, promote vitality naturally. Cordyceps contains all types of nucleosides used in gene regeneration or repairs. It's reported as one kind of bioactive material In this paper, methods were described to determinate nucleosides and their bases in Cordyceps sinensis and cultured Cordyceps mililaris by capillary electrophoresis (CE). And their fingerprints were developed.1 Determination of Nucleosides and their base in Cordyceps sinensis By using adenosine, guanosine and uridine as quality control criteria, extraction process of Cordyceps sinensis had been optimized with the aid of the uniform design U6(63). Influence of pH and the borate concentration was studied on the separation of the standard nucleosides. On the basis, sample separation was optimized using resolution as criteria by the uniform design U6(64) The running buffer of 36 mmol L-1 Na2B4O7-15 mmol L-1 (pH 9.2) was chosen as the optimized condition. 14 0 kV was applied for the separation. 9 active components were assayed They had good linearity (r>0.9992). And the average recoveries of them were more than 93 8% (RSD<5.4) The results were not different obviously between by CE and by HPLC that was regulated in Chinese pharmacopia (ChP).2 Determinnation of Nucleosides and their bases in cultured Cordyceps militaris In order to study the difference of nucleosides content in cultured Cordyceps militaris from different sources, a new method was developed by capillary electrophoresis The running buffer was 40 mmol L-1 borate (adjusted to pH 9.0 with phosphate acid) 16.0 kV was applied for the separation. The wavelength of detector was 254 nm. The average recoveries of 9 active components were more than 93.2% (RSD<4,3%). Quantitative results showed the chromatogram of fruit body was similar to Cordyceps sinensis However, the nucleosides content in cultured Cordyceps mililaris from different sources was changed radically.3. CE / UV Fingerprint Analysis for Cordyceps sinensis and cultured Cordyceps militaris Based on quantitative study, CE / UV fingerprint analysis methods for Cordyceps sinensis and cultured Cordyceps militaris were developed. The methodological evaluation showed that the two sample had both good repeatability and reproducibility. The quality of Cordyceps sinensis might be evaluated by its fingerprint related to the content of nucleosides. And different samples had different CE fingerprints. In this paper, Fingerprints for Cordyceps sinensis and cultured Cordyceps militarishad been developed using the contents of nucleosides as quality control criteria. Themethod might be helpful for the study of Cordyceps sinensis and it's substitution.
