E - selectin is a kind of glucoprotein, belonging to selectin family of adhesion molecules, expressing only on the activated vascular endo-thelial cell. When microenviroment of vascular endothelium changes, it expresses higher. It mediates adhesion of WBC and endothelial cell, after which WBC infiltrate into the matrix, at last endothelium was injured. IR indicates more than normal insulin can execute normal physiologic effect in insulin - sensitive organs. It exists long before diagnose of 2DM,there is causative relationship between IR and 2DM. IR is the key abnormality of metabolic syndrome and is related to the increasing cardiovascular risk of patients. Reviewing documents , many studies focused on the relation between E - selectin and 2 DM or 2 DM with or without vascular complications, resulting that E - selectin levels increased in 2DM patients, also, reports confirmed that in IGT, GDM, essential hypertension, hypertriglyceride patients E - selectin levels increased. The common characteristics of these diseases is IR. In this experiment we have builded IR animal models and discussed the relation between E - selectin and IR.Materials and Methods1. To build IR rat modelSelecting 60 male, adult, Wistar rats (BM<250g) , which were prepared by experimental animal center of CMU. The rats were adap-ting fed for? days before, eating and drinking freely. Dark duration is 12 hours, temperature is (23 ±2)C. Then, the rats were sorted into 3 teams at random, 20 in each team, they were fat - fed team, fructose - fed team and the control team. The calorie intaking is equal a-mong these three teams; 311kJ/d. But the constitute proportion is different , protein, fat, carbohydrate in fat - fed, fructose - fed, normal team were 21% , 59% , 20% ; 21% , 10% , 69% ; 20% , 30% , 50% respectively. The feeds spilled out were complemented on the other day. Fresh feed were made every 3 ~4 days, stored in 4t icebox. Monitoring BG and BM each week. Four weeks later, choosing 5 rats from each team to perform euglycemic clamp test.2. To measure the levels of E - selectin with western - blot method s. This process include material obtaining, centrifuge, protein quantity determining, electrophoresis, transmembrane, hatching with antibody, stain, developing and analysis of E ?selectin levels. Serum FINS, FPG, TG, TC, FFA concentrations were measured with chemi-luminescence immunoassay methods. Endothelium morphology was observed with electron microscope. Data were analyzed with SPSS10. 0 soft wire compact. Means of GIR among clamp teams were valued with one - way ANOVA, means of the variables among material teams also were valued with one ?way ANOVA, linear correlation analysis was performed between E -selectin and TG, TC, FFA, FPG, FINS, HO-MA-%S (log10).Result1. One - way ANOVA showed that E - selectin levels in fat - fed team and fructose - fed team were significantly higher than those in thecontrol team (p <0.01) , in fat - fed team it was higher than that of fructose team ( p < 0.05 ).2. Correlation analysis showed that in fat -fed team and fructose - fed team E - selectin levels correlated significantly with TG, FFA,FINS and HOMA - % S (log10) (r = 0.559, 0.545, 0.526, 0.639, p =0.030, 0.036, 0.044, 0.010; r = 0.575, 0.538, 0.562, 0. 650, p=0.025, 0.038, 0.029, 0.009).3. There were injured phenomena of endothelia observed by the e-lectron microscope.ConclusionExpressed levels of E - selectin correlate with the degree of IR, E - selectin may act as a diagnostic marker of 2 - DM.
