OBJECTIVE: To observe the effect of ShiQuanDaBuTang modification on fe carcinomatous mice who are treated with chemical drug like 5-Fu and discuss it' s mechanism of action.METHODS:Taking out ascites under asepsis which grew well about 9 days as the source of carcinoma and diluting it with salt water which is 2 times more than that. Then injecting 0.2ml diluent into the subcutaneous tissue of the right axilla of mouse. 24 hours after the injection, mice were grouped as control group with tumor, chemotherapy group, ZhenQiFuZheng capsule group, High-dose and low-dose ShiQuanDaBuTang group. After 10 days treatment, we calculated the tumor-supression rate, counted the peripheral blood cells and bone marrow karyocytes, took the weight of the spleen and thoracic gland , observed the contents of IL-2 in serum and NO in spleen and detected expression of Caspase-3 and NF-K B in bone marrow by immunohistochemical method.RESULTS:ShiQuanDaBuTang modification could elevate the tumor-supression rate of 5-Fu on H22 carcinomatous mice, antagonize the toxic and side effects of 5-Fu like the atrophy of the spleen and thoracic gland, the decrease of WBC and bone marrow karyocytes. Also it could increase IL-2 in serum and decrease NO in spleen of carcinomatous mice who are treated with 5-Fu. At the same time, we also observed that it could enhance the expression of NF-K B and suppress the expression of Caspase-3 in bone marrow of carcinomatous mice.CONCLUSION: ShiQuanDaBuTang modification showed marked synergy and detoxication for chemotherapy in H22 carcinomatous mice. Possibly they are related to increasing immunity and regulating apoptosis.
