| Objective:Allogeneic ( C57BL/6j , H-2Kb, BALB/c , H-2Kd ) and homogenous (BALB/c, H-2Kd) mouse models were employed. Observing homing and survival status of transp- lanted cells in mice after bone marrow transplantation with different ways, and relation that homing cells and expression of chemoattractant SDF-1. To investigate the homing law in time of transplanted cells, the affect to homing and survival rate after bone marrow transplantation of different number cells, the role of chemoattractant SDF-1 in hematopoi- etic cells homing. There are three segments in this study.1. The homing law in time of transplanted cells after bone marrow transplantation(DObservetion of homing effective at different endpoint after different number of allogeneic bone marrow cells transplantation by a single injectionExperimental design:Recipient was BALB/c mouse dividing into 4 groups randomly and each was 24 pieces. After 4 hours of total body lethally irradiation of 60Co-Y ray, 5×105, 1×106, 5×106, 1×10 7 different bumber of cells of donor bone marrow cell (C57BL/6j) was transplanted. 6 pieces mouse were to put to death each group respectively at 24 > 48 > 72 > 96 hour after transplantation, detective for (Dkaryote counting of PBC, BMC, SpC, (2)positive cells of donor. Homing status was observed continuously.Results: The positive cells in peripheral blood, bone marrow and spleen detected by flow cytometer was the highest in third day after different number cells transplanted by single injection(P<0.01) .Average homing cells in bone marrow of transplanted groups from first day to fourth day has significant difference(P<0.05) . Similar to above, the homing efficiency from first to fourth day has also a significant difference (P<0.05) .
