Yi Brain Side, Shenshao Of Anti-vascular Dementia Material Basis For Research

Shenshaoyinao Prescription consists of three medicines which are ginseng, red peony root, root of lobed kudzuvine. The effective fractions of Shenshaoyinao Prescription are found to have the effect of anti-vascular dementia. The dissertation divides into three parts to study the material and mechanism of anti-vascular dementia of Shenshaoyinao Prescription.Part one: Literatures and summarizationsDocuments about the present research situation in compound prescription of traditional Chinese medicine which are studied as a whole and the research progresses in the chemical constituents and pharmacological activities of medicines which belong to Shenshaoyinao Prescription, 97 references are cited.Part two: Experiment studies about Shenshaoyinao Prescription.One, the effects of anti-vascular dementia from Shenshaoyinao Prescription are extracted and isolated. 10 compounds are isolated with silica gel, polyamide, sephadex gel chromatography and 8 compounds are identified with UV,FAB-MS,1HNMR,13CNMR spectra. They are one isoflavanones(daidzein), six isoflavanonal glycosides(4′-O-methyl-daidzin, 4′,7-diglucoside, genistin, puerarin, PG-3, daidzein 8-C-Apiosyl(1â†'6)glucoside), one monoterpenoidal glycosides(paeoniflorin). All of the 8 compounds are firstly obtained from Shenshaoyinao Prescription. Two, HPLC figs of the effects of anti-vascular dementia from Shenshaoyinao Prescription are established primarily. By analyzing and comparing, 28 chromatographic major peaks are found in the HPLC figs of the effects of anti-vascular dementia from Shenshaoyinao Prescription and the ascriptions of the peaks are validated in the HPLC figs of corresponding and negative corresponding part to every single medicine. 10 of 28 peaks are confirmed as 4′,7-diglucoside(2), puerarin(4), PG-3(5), paeoniflorin(6), daidzein8-C-Apiosyl(1â†'6)glucoside(9),4′-O-methyl-daidzin (14), ginsenoside Rg1(15), ginsenoside Re(15), daidzein (17), ginsenoside Rb1(19).Three, the total saponins contents in the effects of anti-vascular dementia and the corresponding part of ginseng and red peony root are determined. They are 16.9%, 41.1%, 35.9% respectively. The linear range of ginsenoside Rg1 is 0.072-0.360 mg, the correlation cosfficients are 0.9995. The contents of index compounds including puerarin, paeoniflorin and daidzein are determined by HPLC. The calibration curves are linear at range of 0.4-2.4μg (for puerarin, r=0.9996), 0.171-0.855μg (for paeoniflorin, r=0.9996), 0.17-0.85μg (for daidzein, r=0.9997), respectively. Theaverage recoveries are 103.4%(RSD 1.16%,n=5), 96.9%(RSD 2.39%,n=5), 101.3%(RSD 2.04%,n=5), respectively.Part three: analysis and discussion of experiments.
