ObjectiveThrough observing the effect of Qiang Gu capsules (QG) on bone metabolism of rats with osteoporosis(OP), we explore the acting mechanism and curative effect of QG on primary osteoporosis(POP) caused by deficiency of liver and kidney ,deepen the recognition on TCM pathogenesis of POP,and provide an experimental basis for clinical medicating and further exploitation.MethodsThe sixty-six femal 3-4 month's Wistar rats were randomly divided into six groups: normal control group "normal group"(A), osteoporosis model control group "model group"(B),nylestriol -treated group(C), QG in low dosage (D),QG in middle dosage(E) and QG in high dosage (F). Each group was 11 rats,measuring these rats weights and temperatures before beginning of the experiment. Excepting the normal group ,the rats of each group were made bilaterally ovariectomized . Three days later after the operation ,the living rats were again randomly divided into five groups ,and each group was 9-11 rats .The rats of the normal group were feeded in the same conditions and not specially delt with. The rats of each group had been given different medicines by gastrogavage for 12 weeks. All rats were injected tetracycline(25ml/kg) into their abdomen and made double sign of bone separately at the day before ten days and two days of the experiment end. At the end of the experiment, we separately measured weights, temperatures of all rats and took blood, bones for tests ,etc.Results1 The femur dry weight, bone mineral density(BMD), calcium in bone determination and some parameters of shankbone histom orphometry in the model group at the experiment were significantly lower than thoses in the normal group .which showed the osteoporosis rat model established.2 The animals in each group didn't show any bad reactions .The weights and temperatures between the six groups didn't present statistical difference.All rats grew and actived normally,whose hair didn't lose.3 The femur dry weight,BMD, and calcium in bone determination in each dosage group were significantly higher than thoses in the model group(P<0.05,or P<0.01),which were similar to the nylestriol -treated group.4 The major indexs which reflected the level of bone mass,i.e., the shankbone's Tbv, percentage of Tbv, average bone trabecular width ,total bone trabecular length, length of double sign of bone trabecula and average width of double sign of bone trabecula in each dosage group were significantly higher than thoses in the model group (P<0.01).The average bone trabecular gap width were significantly lower than thoses in the model group (P<0.01).5 The level of bone gla protein(BGP), bone mineralization rate(BMR) and acidity pectic amylases which reflected bone formation, in each dosage group weresignificantly higher than thoses in the model group (P<0.01).The level of BGP and acidity pectic amylases in the middle and high dosage groups were significantly higher than thoses in the nylestriol -treated group(P<0.01).6 The femur biomechanic parameters in each dosage group were significantly higher than thoses in the model group(P<0.05 ~ 0.01), similar to the nylestriol -treated group.7 The weights of uterus in middle dosage group and nylestriol -treated group were significantly higher than thoses in the model group (P<0.05 ~ 0.01).8 The effect of QG improved following increase of it's dosage.ConclusionThe results of the experiment indicated that above observed indexes were significantly better than thoses in the model group and that some indexes were better than the nylestriol -treated group after the osteoporosis rats were treated by QG From microcosmic index of serum chemistry test .because QG capsules activated osteoblast to produce bone matrix through increasing the level of BGP and improved bone matrix's materialization ,it could promote bone formation. From relevant indexes of bone histom orphometry, QG capsules could inhibit bone asorption ,promote the increase of bond mass, improve microstructure of bone of rats with OP and increase acid...
