Objective To probe into the effects of electroacupuncture on immediately early gene c-fos of cortex in rats during reperfusion after focal cerebral ischemia, and verify the relationship between c-fos gene and mechanism EA unti-injury on brain ischemia. Methods using double- labeled technique to make model of MCAO (middle cerebral artery occluded) rats, the experiment was random divided into seven groups, simulate operation group, 90min. cerebral ischemia and reperfusion Ih 3h 9h groups,90min. cerebral ischemia and reperfusion with electroacupuncture 1h 3h 9h groups. We observed the number of positive cell be expressed by c-fos protein in the cerebral cortex of ischemic side of rat and the change of the volume of brain ischemic infarct and the score of nerve function defect with means of immunohistochemical (SABC) and TTC staining. Results It was found that there are more positive c-fos protein expressed cells and smaller volume of brain ischemic infarct and lower scores of nerve function defect than that in contrast groups, and also have outstanding difference (p<0. 01). Conclusion these results indicated that EA may increase expression of c-fos gene of cortex in the rat during reperfusion after focal cerebral ischemia, and it would be one of the mechanisms of EA anti-injury cerebral ischemia.
