Sugar Ann Fluid Diabetic Gastroparesis Plasma Endothelin And Nitric Oxide

Objective: To study the effects of Chinese herbs TangAnYe(TAY) on plasma endothelin(ET) and nitric oxide(NO) in gastric tissues in experimental animals, we established the rat model of diabetic gastroparesis(DGP).This research tried to study and analyse the mechanism of preventing and treating DGP.Methods: Except those of the normal control group, the other rats were used as pathology models of DGP. They were induced by being injected streptozocin(STZ),50mg/kg and their blood sugars were higher than 16.65 mmol/L.64 Wistar diabetic rats were randomly divided into four groups: pathology model group, Cisapride treatment group ,TAY intragastric administration treatment group, TAY coloclysis treatment group. Each group had 16 rats. Another 16 healthy rats were chosen as the control group. The control group was fed regularly on standard rat food. The other four groups were fed on high calorie food (high sugar and high fat)irregularly ,eating in the morning in odd-number days and in the afternoon in even-numbered days. All the rats drank freely.During the experiment periods, the appetite, water volume, urine volume, stool character should be observed, and weigh the rats, test the urine sugar once a week. Each group was perfused stomach or coloclystered with drugs during establishing the model. The control group and the pathology model one were perfused stomach with saline,10ml/kg.TAY intragastric administration treatment group 10ml/kg(5.8g/kg),TAY coloclysis treatment group 20ml/kg (11.6g/kg) and Cisapride treatment group 10ml/kg(2.0mg/kg),once a day,lasted 6 weeks. Recorded the weight and the blood sugar on unempty stomach. The day before the last day, fasting 24 hours, 2 hours before last feeding, the rats were perfused stomach with liquid-solid paste.30 minutes later, sampled blood in rats' eyeball to measure blood sugar and plasma ET. Immediately the rats were put to death. Then got out the rats' stomach to measure the emptying rate. At last cut some tissues in gastric antrum to measure NO.Results:TAY could improve the symptoms of the DGP rats significantly,decrease the levels of the blood glucose. The effects were better than that of the pathology model group(P<0.01 ) .TAY was also able to promote gastric emptying. Compared with the Cisapride treatment group, there was no significant difference(P > 0.05).TAY was able to decrease the levels of plasma ET and increase the levels of NO in gastric tissues. The effects were better than the pathology model group and theCisapride treatment group (P < 0.01 ) .Conclusion: TAY can ameliorate the symptoms of DGP rats, decrease blood glucose level effectively, improve the blood lipid and blood viscosity, promote gastric emptying, decrease the levels of plasma ET and increase the levels of NO in gastric tissues. These findings suggested that TAY may have a value in preventing and treating of DGP.
