ObjectiveThe diabetic rats were induced by Ip of STZ and were fed with high calorie feed irregularly. To study the effect of Tanganye (TAY) on Diabetic Autonomic Neuropathy by irrigating enema in stomach and in intestine, and try to discuss the preventive and therapeutic effect of TAY on Diabetic gastroparesis (DGP). To offer the scientific basis for TAY' clinic use and exploitation. Provide an effectively preventing and treating drug for patient of this disease.MethodsWe chose some male rats of Wistar (weight of 180?10g), and fed them in lab for 1 week to let them adapt the condition. The rats were induced with STZ by IP ( 50mg/kg) . Then we chose the rats that their blood glucose were higher than 16. 7mmol/L, divided these rats into four groups randomly : model group(group I ), Cisapride group (group II ), TAY irrigated in stomach group (group III) ,TAY retention enema in intestine group (group IV). 16 rats each group. Further more we chose another 16 healthy rats as control group (group V). The group V were fed regularly on standard rats feed . The other four groups were fed on high calorie feed irregularly, eating on morning in even-numbered days and eating on afternoon in odd-numbered days. Al 1 the rats drunk water freely. During the experiment period, the appetite, water volume, urine volume, stool character should be observed, and weighed the rats every week. Each group was irrigated with drugs during establishing the model. The group land Vwere irrigated stomach with saline 10ml/kg. Group III with TAY 10ml/kg (5.8g/kg) . Group IV with TAY 20ml/kg (11. 6g/kg), GroupII with Cisapride lOml/kg (2.0mg/kg). once per day, lasted 6 weeks. Recorded the weight and the blood glucose in the day before the last day. On the last day executed all rats after they were irrigated semisolid paste and were collected sample blood. Measured the gastric remnant rate, content of sorbitol in red cell, and observed the mophorlogic change of the pneumogastric nerve.ResultsComparing with group V,group I 's gastric remnant rate and the level of the blood glucose was markedly higher. (P<0. 01) .After 6 weeks' treatment, each therapeutic groups' gastric remnant rate was lower than group I ( P<0. 05, P<0. 01). But there was no significant difference between TAY therapeutic groups and Cisapride therapeutic group. Comparing with group I , the content of sorbitol in red cell waslower markedly ( P<0.01 ) in group III and IV ,but group II was no different(P>0. 05 ). Group III and group IV ' mophorlogic nerves were more than group I and II ( P<0. 01).ConclusionsThe experiment results show: (1)the method of using STZ and high calorie feed with irregular feeding could make the model of DGP; (2)the therapeutic effect of TAY irrigated in intestine is better than TAY irrigated in stomach on improving the empty of stomach, the same as Cisapride group; (3)the treatment with TAY could get better therapeutic effects on DGP by improving damnification of nerves; (4)TAY can decrease the level of the blood glucose, blood lipid and blood viscosity.
