Objective:To study preventive and therapeutic effects of WuWeiShu on disorder of Pituitary-Adrenal System during Glucocorticoid(GC) treating Nephrotic Syndrome (NS) rats. Methods:Sixty male Wistar rats were selected and randomly divided into two groups. 12 rats were used as normal group, and the remaining rats were given intravenous injection of Adriamycin (0.6mg/100g) to establish model of NS. The model of NS rats were divided into four groups: the model group(n=12), WuWeiShu group(n=12) treated with WuWeiShu alone, prednisone group(n=12) treated with prednisone alone, WuWeiShu and prednisone group(n=12) treated with WuWeiShu combined with prednisone for 28 days respectively. The general condition of rats, 24 hour urine protein quantity(24HUPQ), serum albumin, total protein, creatinine, urea nitrogen, triglyceride, cholesterol, cortisol, plasma adreno-corticotropin hormone (ACTH) and pathological changes of renal tissues were observed after treatment. Result:24HUPQ increased significantly 7 days after injection of Adriamycin. After 28 days treatment, all treatment groups showed decreasing in 24HUPQ, triglyceride, cholesterol(P < 0.05, P < 0.01)and increasing serum albumin and total protein (P < 0.01)compared with the model group. The results of ACTH and cortisol showed: prednisone group decreased significantly(P < 0.01), while WuWeiShu group increased significantly(P < 0.01)compared with the model group. And WuWeiShu and prednisone group increased significantly (P < 0.01,versus the other treatment groups).Pathological studies showed obvious inosculation of podocytes without glomercular base membrance thickening and mesangial matrix proliferating in the model group, while partial inosculation of podocytes in all treatment groups. Conclusion:The results showed:(1)the methods of intravenous injection of Adriamycin could induce successfully the NS model;(2)all treatment groups could get the therapeutic effects;(3)the therapeutic effects of WuWeiShu and prednisone group were better than the others; (4) WuWeiShu could regulate disorder of Pituitary-Adrenal System during GC treating NS. WuWeiShu could decrease the side-effects and increase the therapeutic effects of GC by adjusting Pituitary-Adrenal System.
