Matrine is a alkaloid which is abdundant in sophora Flavescens Ait S.Qlopecuroioles L, S. snbprostratachunat T.Chon etc, which has inhibition on the experimental tumor. It is reported that Matrine can lighten the symptoms, lengthen survival period, does not destroy the production of the normal leucocyte, can even raise the number of leucocyte, improve organism resistance used in treating various kinds of later cancer, this is difficult to many chemotherapy medicines. Liposomes is used as a carrier of anticancer drugs extensively in recent years, it can improve chemotherapy target tropism of medicine to a certain extent, reduce chemotherapy medicines malicious effect of medicine by a large margin, raise chemotherapy treatment index of medicine from several ways. Compared with other forms of an anticancer drug at present, liposomes has unique advantages, it can reduce toxicity and continue keeping cells' poisonous function. Because it can improve stability target tropism and long result of medicine and reduce dosage, etc. We selected liposomes to entrap matrine for formulating high-efficient natural medicine against cancer.Firstly, we studied the membrane material purchased in the market by oxidizing index method, the result showed that the soybean phosphatide fulfiled the requirement. The liposomes were prepared by the following methods: TFV, REV(reverse-phase evaporation vesicle method), two emulsification vesicle method, Kirby method pH gradient vesicle method, sulphuric acid ammonium gradient vesicle method, FTV, freeze-drying vesicle method, etc. To determine the encapsulation of liposomes we used high-efficient capillary electrophoresis. The result indicated the encapsulation of liposomes prepared by REV was the highest. To grope the optimal technical condition, we used orthogonal text. At last, the highest encapsulation was 50.48, we adopted the optimal technical condition combined freeze-drying method to make liposomes, then examined encapsulation, the encapsulation slightly droped. Scanning Electron Microscopy and Vision analysis instrument were used to observe its morphology and vesicle size respectively. Most liposomes were single-room spherical liposomes, the diameters of liposomes prepared by REV were 0.186-3.537μm, mostly smaller than 0.332μm, After freeze-drying the diameters of liposomes were mostly smaller than 3.15μm and average 3.497μm. Pharmacology experiment set up high middle, low three dosages group to observe the three dosages group resist the tumour effect to little mouse S-180, the result showed, middle dosage was effectivest. Then made the tumour result price research with middle dosage. Results proved that matrine liposomes was obviously stronger than matrine to little mouse's immune organs, function of H-22, EAC of mouse, lower than CTX, but there was no significant difference. Matrine liposomes was obviously higher than CTX to the little mouse H-22 lymphocyte conversion ratio, higher than matrine. This text had offered certain theoretical research for the study of matrine liposomes.
