Objective: Nephrin and Podocin are identified as crucial proteins ofthe slit diaphragm, a specialized cell-cell contact between interdigitating foot processes of the podocyte. We observed influence of traditional Chinese medical prescriptions on expression and distribution of the two proteins on podocyte of adriamycin-induced nephrotic syndrome rats and investigated therapeutic effect of traditional Chinese medical prescriptions on nephritic syndrome and its potential mechanism.Methods: SD rats were randomly divided into six groups: normalgroup, model control group, traditional Chinese medical prescription 1 group (TCM1), traditional Chinese medical prescription 2 group (TCM2), traditional Chinese medical prescription 3 group (TCM3) and prednisone group. Indirect immunofluence staining technique was used to detect expression and distribution of Nephrin and Podocin on podocyte of glomeruli.Results: (1) Antibodies against the cytoplasmic domain of Nephrin and Podocin showed a typical linear epithelial-like reactivity within glomeruli in rats' kidneys of normal group. (2) In ADM-treated groups, daily urine protein rose at day 10 and reached peak at day 30; The quantitative expression of Nephrin and Podocin was reduced in comparison with normal group and a patchy and less intense pattern of reactivity of fluorescence was seen. (3) After administration, dailyurine protein in three TCM groups were less than model control group, especially in TCM2 group. In addition, quantitative expression of Nephrin and Podocin in three TCM groups was significantly higher than that of model control group.Conclusion: Daily urine protein excretion of adriamycin-inducednephrotic syndrome rats could be reduced effectively by traditional Chinese medical prescriptions. To modulate expression of Nephrin and Podocin on podocyte within glomeruli is likely to be one of potential mechanisms of this effect.
