Effect Of Qingre Zhixue Granule And Glucoside Of Tripterygium Wilfordii On The Expression Of Nephrin,Podocin And Podocalyxin In The Injured Model Of The Podocyte Of Henoch-Schonlein Purpura Nephritis

Objective: To explore the drugs' effects in the treatment of Hen?ch-Sch?nlein purpura nephritis through the observation of Qingre Zhixue Granules,Tripterygium wilfordii multi glycoside(GTW)and other drugs on the expression of nephrin?podocin and podocalyxin proteins of HSPN's podocyte injury model of glomerular..Methods: Mouse mesangial cells and mouse podocytes are cultured in vitro,separation and purification of the m Ig A1 in serum in children with Hen?ch-Sch?nlein purpura nephritis,thermal polymerizated into the aggregated Ig A1(a Ig A1),a Ig A1 and mouse mesangial cells were cultured together for 48 hours as the culture medium of podocyte 's injury model,at the same time,adding medicine Qingre Zhixue Granules,GTW and GTW combined with Qingre Zhixue Granules,hormone combined with heparin,blank serum and fetal bovine serum.Collect podocytes after stimulated for 24 hours(24h)and 48 hours(48h),testing the expression of podocytes related protein by Western Blot and immunofluorescence methods.Results: ??24h and 48 h later,nephrin,podocin and podocalyxin proteins' expression were detected by the Western Blot method,the results show that the normal control group was the highest,the model group was the lowest(P < 0.01):(1)24h nephrin protein's expression indicated that the GTW group?GTW+Qingre Zhixue Granule group and hormone+heparin group's nephrin protein was significantly increased compared with the model group(P<0.01),among the three groups,there was no difference(P>0.05),there's no significant differences between the Qingre Zhixue recipe group and model group(P>0.05);48h later,the expression of GTW+Qingre Zhixue Granule group was higher than hormone+heparin group(P<0.01).(2)24h podocin protein's expression indicated that the GTW group?the GTW+Qingre Zhixue Granule group and the hormone+heparin group's podocin protein was increased compared with the model group(P<0.05),hormone+heparin group's protein was significantly higher than those in the GTW group's,but with GTW+Qingre Zhixue Granule group there's no difference(P>0.05),the expression of Qingre Zhixue Granule group had no difference compared with the model group(P>0.05);48h later,the GTW group?GTW+Qingre Zhixue Granule group and hormone+heparin group's podocin protein was significantly increased compared with the model group and the Qingre Zhixue Granule group(P<0.01),the expression of Qingre Zhixue Granule group had no difference compared with the model group(P>0.05),proteins' expression of hormone+heparin group increased significantly than GTW group(P<0.01),but there was no difference compared with GTW+Qingre Zhixue Granule group(P>0.05).(3)24h podocalyxin protein's expression indicated that that the GTW group ? the GTW+Qingre Zhixue Granule group and hormone+heparin group's podocalyxin protein was increased compared with the model group(P<0.05),the expression of Qingre Zhixue Granule group had no difference compared with the model group(P>0.05),the expression of hormone+heparin group was higher than GTW+Qingre Zhixue Granule group's(P < 0.05);48h later,the GTW group?the GTW+Qingre Zhixue Granule group and the hormone+heparin group's podocin protein was increased compared with the model group(P<0.01),but there's no significant difference between three groups(P >0.05).??Detection of podocyte's structure protein nephrin,podocin and podocalyxin's positioning expression by IF showed: neprin expressed in podocyte cytoplasm in the blank group,podocin expressed in podocyte membrane,podocalyxin in the cytoplasm of cells was granular expression;In model group,nephrin's expression decreased significantly,there was no expression of podocin and podocalyxin;Each drug could enhance the expression of podocyte injury model's protein expression.Stimulation of 48 h compared with 24 h,the cells' number decreased,cell morphology is not complete,part of the cells were like broken floc.Conclusions:1.GTW,GTW+Qingre Zhixue Granule and hormone+heparin have podocyte protective effect.2.GTW+Qingre Zhixue Granule may be better than hormone+heparin in protecting the nephrin protein of podocyte;Hormone+heparin in the protection of podocin may be better than the single use of GTW,GTW+ Qingre Zhixue Granule can enhance the effect.Hormone+heparin in the protection of podocalyxin is faster than GTW and GTW+Qingre Zhixue Granule,however,extending the time there's no difference in efficacy.
