Effect Of Yi Shen Capsule On The Expression Of Nephrin And Podocin In Mouse Podocytes Under High Glucose Environment

Objective:The improved method of serum pharmacology was used in this study,After oral administration,the serum was taken as the medicine source,and the exogenous high glucose as the stimulus was studied,To investigate the effect of Yishen Capsule on the expression of nephrin and podocin in cultured mouse cells,In order to provide a theoretical basis for the Yishen Capsule in treatment of diabetic nephropathy.Methods:Cultured mouse podocytes strains induced differentiation of mature after divided into 7groups:normal group,high glucose group,isotonic group, Yishen low dose group, middle dose group, high dose group,the benazepril group,in accordance with the above group after cultured for 48 h,with CCK-8 method detection cell proliferation,analysis of expression of nephrin,podocin using immune fluorescence semi quantitative method,with Western blot detection of nephrin, podocin protein content.Results:CCK-8 methed showed that podocytes in all groups after 48 hours of culture, compared with the normal group, the proliferation of foot cells in high glucose group decreased significantly(P<0.01), the difference is statistically significant. compared to high glucose,Yishen high dose group, benazepril group foot cell proliferation increased significantly(P<0.01), and the difference was statistically significant.Western blot showed that podocytes in all groups after 48 hours of culture, compared with the normal control group, high glucose group of nephrin and podocin protein expression decreased significantly(P<0.01), the difference is statistically significant;group and high glucose compared to Yishen high dose group and benazepril group of nephrin and podocin protein expression increased significantly(P<0.01), the difference is statistically significant.
