In order to clarify genetic toxicological effect of heroin on drug addict, the frequency of sister chromatid exchange (SCE). micronucleus (MN) and Ag-staining nucleolar organizing region( Ag-NOR) of heroin addict was examined in this paper.In chapter 1, to detect damage degree of DNA in heroin abuse, the sister chromatid exchange frequency was counted in the sample of 33 patients with SCE different staining method. In the same time, we detected 30 normal controls. The results show that the SCE frequency of lymphocyte in patients was higher than that of the controls, P<0.01. Conclusion: Heroin is able to increase the damage degree of DNA of the human body.In chapter 2, to study the effect of heroin on the lymphocyte micronucleus rate of male drug addicts, the lymphocyte micronucleus rate of 40 male drug addicts was tested and compared with micronucleus rate of 30 normal men. The results show that the lymphocyte micronucleus rate of male drug addicts was higher than that of normal men and the difference was obvious (P<0.05). Conclusion: The heroin may cause the increase in lymphocyte micronucleus rate in male. The heroin can damage the chromosome. It may be the mutagenic material.In chapter 3, to study the effect of heroin on the activity of rRNA genes in NOR. The NOR and acrocentric association were studied in 30 male drug addicts and 30 normal men by the method of Ag-NOR. The results show that the distribution of the Ag-NOR and Ag-AA in the drug addict group and control was random. The frequency of the Ag-NOR in drug addict group was lower than that of control. There were no significant differences in the frequency of Ag-AA between the two groups. Conclusion: The male drug addicts of over two years showed that heroin have certain degree of restraint to the activity of rRNA genes.
