Different Route Of Administration Of Ulcerative Colitis Immune And Repair Mechanisms, Comparative Study

Background and ObjectsWith the development of living standard and with the obviously change of diet the morbidity of Ulcerative Colitis has appeared a trendency of increasing .It is a kind of nonspecific inflammatory disease of rectum and colon .Since it's difficult to heal UC and it has a large chance to recur again ,WHO defined it as a disease which is difficult to cure. And it also became a hotspot in the field of Gastroenterology .Under the guidance of ancient datum and the experiments of famous doctors Traditional Chinese Medicine shows superiorities in clinical studies .The peculiarity of TCM is his lower proportion of relapses ,doubtless effects of therapy and less toxicity or side-effects .Now it has make a lot of progresses and innovations in theories ,methods ,prescriptions and herbs of TCM .There are three main methods to treat Ulcerative Colitis .We can use medicine ,make a clysters ,or join them together .All the way gain good effects in clinic .This article makes a summary about the evolutional status of therapies of TCM .And sum up the mechanism which promote the growing of UC .In the first part we talk about the medicine ,clysters ,and both of them separately .In the second part we hope to find the different effects on the mechanism by different way of medicine giving .Finally choose the best way which is based on the results . MethodsThe UC rat model was induced by TNBS/ethanol intermixture . In addition we fed fifteen normal rats as contrast .All the rats were divide into 5 groups .Normal group were given distilled water;model group were given distilled water also ;Olsalazine sodium group were treat by drinking solution of Olsalazine ; Kui Jieyin drinking group were treat by drinking Kui Jieyin ; Kui Jieyin clysters group were treat by clyster of Kui Jieyin .After interfere by medicine we observe following changes on different way of treating in different period . (1) Observe the effects on weight and general status after different way of treating . (2) Observe the effects on pathological changes on the mucous membrane of rectumand colon after different way of treating . (3)Check-up the effects on NO concentration in blood serum after different way oftreating . (4)Check-up the effects on inflammatory cytokine's concentration in blood serumafter different way of treating by ELISA reagent box . (5)Check-up the effects on the expression of EGF and TNF-a on the mucousmembrane of rectum and colon after different way of treating by ELISAreagent box . ResultsHaving the medicine in different way will produce different effects on the mechanism of immune system .But it can't be described just as having medicine in oral way can only adjust the state of holistic immune system and clyster can only have effects on the part it has contact. (1)The effects on weight and general status after different way of treating :First week the weight drop off in all groups except normal group. Kui Jieyin can partly increase the weight when eating and clyster .After second week the rat's weight in Kui Jieyin groups have no difference with normal group. (2)The effects on pathological changes on the mucous membrane of rectum and colon after different way of treating :Prophase feeding by Kui Jieyin can decrease the number of necrotic tissue and ulcer as well as vasculitis .In metaphase decrease the chronic soakage of inflammatory cell ,destroy the epidermis and adenalgia, accelerate the proliferation of granulation .Anaphase it can reduce the tardive ulcer .While clystering by Kui Jieyin have the similar effects at the same phase .And in addition it can limit the area of inflammation but can't alleviate the level of inflammation and tardive ulcer.(3)The effects of different treating methods on the concentration of IL-4 and IFN-γ, as well as ratio of IFN- γ /IL-4, in DC rats serum:The different treating methods have similar effects on the immune regulation to decrease IFN- Y and increase IL-4, as to adjust the ratio of IFN- Y /IL-4 and inhibit the over reaction of inflammation.
