Objective: Through cultivated myocardial cell in vitro rat, to explore RB, RS and their compatibility extract antivirus and the protective function by CVB3m infection on different administration and time and to initially clarify the mechanism of their therapeutic effect.Methods:1. Principal drug and ministerial drug and their compatibility serum were got by serum pharmacology.2. We measured maximal no virulence dose of the medicine serums and toxicity of CVB3m.3. The myocardial cells were cultivated in vitro.4. The medicine serums had affected on toxicity of CVB3m in vitro.5. We observed morphological variety , pathological changes and the percentage of pulsates of the myocardial cells by CVB3m infection after 3, 5 days.6. Every protect rate of the myocardial cell of every medicine serum dissimilarity to infect with CVB3m for the medicine time by MTT method.7. We determined lactate dehydrogenase and aspartate aminotransferase release in respectively supernatants on different administration and time , contrasting the protective effect of every medicine serums on myocardial cell by CVB3m infection.8. Reverse transcription polymerse chain reation (RT-PCR) was used to exam expression of CVB3m RNA in myocardial cell administration on the differet time, investigated every medince serum has the different effect on CVB3mRNA replication.Results:1. RS group might have affect on directly kill CVB3m in vitro.2. We observed morphological variety , pathological changes and the percentage of pulsates of the myocardial cells by CVB3m infection after 3, 5 days. The effect of RB and compatibility group were better than RS extract in advance administration. The difference RB and compatibility extract wasn't significant. The effect of RS and compatibility extract were better than RB extract after administration.3. The three kinds of medince serums all could protect myocardial cell by CVB3minfection .The effect of RB and compatibility group were better than RS group in advance. The difference RB and compatibility group wasn't significant.Using MTT showed every medicine serums can protect rate of myocardial cell was significant.4. The three kinds of medince serums all could degrade lactate dehydrogenase and aspartate aminotransferase release in respectively supernatants on different administration and time , RS and compatibility group was more significant than RS.5. The CVB3m RNA expression in myocardial cell were by RT-PCR explored the effect of antivirus , the one of RB and compatibility extract is more significant than RS.Conclusion:1. RS group might have affect on directly kill CVB3m in vitro.2. The three kinds of medince serums all could protect myocardial cell on degree before CVB3m infection . The effect of RS and compatibility extracts was significant , and might inhibit CVB3m adsorption and infection, so might protect the myocardial cells by CVB3m infection.3. The three kinds of medince serums all could protect myocardial cell by CVB3m infection on degree after administration. The serum of RS and compatibility extracts were obviously inhibit CVB3mRNA replication and the cell by CVB3m infection had obviously been protected.4. Compatibility serum had antivirus and protection in vitro , but no specifically dominance.
