This research extract four fall-blood sugar effective extracts (ginsenoside,ophiopogonpolysaccharide, astragalus membranaceus flavone, schisandra chinensis lignanoid)were extracted respectively from four traditional Chinese medicine in the treatment of diabetes(red ginseng, radix ophiopogonis, astragalus, schisandra chineisis), and investigate extractiontechnology of effective material with decreasing sugar. By combining traditional Chinesemedicine and modern biological technology, the research construct type2diabetes animalmodels, and the related biochemical indexes such as the serum insulin, fasting blood sugar,serum creatinine, triglycerides, total cholesterol was selected to screen compatibility group,which has potential to reduce blood glucose and prevent the complications of diabetes.Optimizing extraction technology of the four active substance by orthogonal test.Ginsenosides were extracted with ethanol, investigation factors were including ethanolconcentration, dosage liquor ratio, extraction times and extraction time, it was concluded that thebest extraction technology was10times the amount of70%ethanol circumfluence extraction2times, each time120min,the total content of Rg1and Re was3.530mg/g; Ophiopogonpolysa-ccharide was extracted with water, consideration factors was the dosage liquor ratio, extractiontimes, extraction time and sinking alcohol concentration, it was showed that the best extractiontechnology was10times the amount of water with extracted three times, each time60min,sinking alcohol concentration was90%,the content of Ophiopogonpolysaccharide was18.18%;With ethanol extracting astragalus membranaceus flavone, factors including ethanolconcentration, dosage liquor ratio, extraction times and extraction time, it was concluded that thebest extraction technology was10times the amount of70%ethanol circumfluence extraction2times, each time60min,the content of astragalus membranaceus flavones was measured1.739mg/g; Schisandra chinensis lignanoid was extracted with ethanol, examine factors wereethanol concentration, dosage liquor ratio, extracting times and extracting time, it was concluded that the best extraction process was8times the amount of90%ethanol circumfluence extractiononce, extraction time1.5h, the content of Schisandra chinensis lignanoid was2.447mg/g.This research established type2diabetic rats model with high sugar and high fat diet plussmall dose of Streptozotocin (STZ:35mg/kg) peritoneal injection,90male SD rats wererandomly divided into normal control group and high-sugar high-fat feeding group, after insulinresistance, high-sugar and high-fat group was injected with STZ to induce type2diabetes model,normal control group rats injected with the same dose, citric acid-citrate sodium buffer solution,finally rate of success model was76.2%,64in all.Through analysis and comparison fasting bloodglucose of diabetes rats, making each group of rats with closing blood glucose value, anddiabetes rats were divided into8groups (each group of8) respectively, intragastricadministration with physiological saline, metformin enteric-coated metformin hydrochloride (0.2g/kg), Tang-maikang granule (1.58g/kg), the compatibility of one (0.5:1:2:1), the compatibilityof two (0.75:1:2:1), the compatibility of three (1:1:2:1), compatibility of four (1.25:1:2:1),compatibility five (1.5:1:2:1), normal control group by gastric saline solution. Executed rats after3weeks medication, taking serum, kidney specimens. Using biochemical, ELISA method todetect the serum insulin, serum creatinine, blood glucose, triglycerides and total cholesterollevels.Experimental results showed that the compatibility of four and compatibility of five hadbetter effect than other compatibility group in hypoglycemic, both groups had no statisticalsignificance with Tang-maikang (P4=0.372>0.05, P5=0.752>0.05), two groups ofglucose-lowering rate (21.6%,21.4%) were similar with Tang-maikang group (20.2%); ISI, IRIof compatibility four compared with Tang-maikang group had statistical significance (P <0.05), and it better than Tang-maikang. Compatibility of four can effectively reduce the totalcholesterol and triglyceride level, and lipid-lowering effect was better than the positive controlgroup metformin (P <0.05). curative effect of compatibility five was better than that ofmetformin positive control group on serum creatinine (P <0.01), and had significantdifference.Compatibility four and five of TCM showed good curative effect,the ratio ofastragalus membranaceus flavones in the compatibility had a positive correlation with the effect.The research results of serum insulin, insulin sensitive index, glucose tolerance test,blood lipids and other related parameters of diabetes showed that the compatibility of traditional Chinese medicine combination can adjust insulin resistance of damaged rats, can increase insulinsensitivity of target organ, improve the high serum insulin hematic disease, effectively reduce thecontent of blood sugar in the body. And had obvious regulation effect on blood lipid metabolicdisorder of diabetes rats, can prevent hyperlipidemia complications. Comprehensive analysis, thecompatibility of four and five have the better effect of falling blood sugar than Tang-maikanggroup, compatibility of four lipid-lowering effect is superior to metformin group, compatibilityof five has the better curative effect in serum creatinine than metformin group.
