Mechanistic Studies Of Liver Detoxification And Collaterals Treatment Of Experimental Rat Hepatoma

Objective Probing into the Traditional Chinese Medical Pathologensis of primary carcinoma of liver.To study the relationship of cell proliferation and apoptosis with primary carcinoma of liver , clarify the mechanism of qing-gan-jie-du-tong-luo prescription restraining primary carcinoma of liver.Methods ①Wistar male rats were divided into 5 groups :normal group, model group, western medicine group, qing-gan-jie-du-tong-luo prescription low dose group and high dose group.Rats were subcutaneous inoculated SMCR-86 neoplasia mass testosterone propionate for 10 days.②Observe the changes of liver weight , prostate exponent , histology under light microscope of each group. ③ Take the radioimmunochemistry to measure the aminotransferase and aspartate aminotransferase content of liver constitution. ④ Use the method of immunohistochemistry to detect the expression of alpha fetoprotein,proliferating cell nuclear antige in each liver constitution.⑤Taking the flow cytometry to measure the cell apoptosis rates of liver constitution.Results ①The liver weight of model rats increased significantly ( P<0.01 ), but body weight decreased , qing-gan-jie-du-tong-luo prescription can restaint the changes. ② In histology the model rats have obvious anomaly , qing-gan-jie-du-tong-luo prescription can relieve the anomaly.③The aminotransferase and aspartate aminotransferase content of liver in model rats increased greatly ( P<0.01 ) , qing-gan-jie-du-tong-luo prescription can low the level. ④ The expression of alpha fetoprotein,proliferating cell nuclear antige increased significantly ( P<0.01 ), qing-gan-jie-du-tong-luo prescription can low the PCNA,AFP expression. ⑤ The cell aptosis rate in model liver cells decreased significantly , qing-gan-jie-du-tong-luo prescription can increase the rate.Conclusion The study has suggested that the "qing-gan-jie-du-tong-luo" is the fundmental treatment principl of primary carcinoma of liver. By the experimental study , we detected the aminotransferase and aspartate aminotransferase content of liver in model rats increased greatly, the expression FAP and PCNA increased in model group,but the cell aptosis rate in model liver cells decreased significantly.The results suggest these result in primary carcinoma of liver. Qing-gan-jie-du-tong-luo prescription can significantly control weight of rats' liver , we presume qing-gan-jie-du-tong-luo prescription can regulate the cell factors , induce the cell apoptosis of liver.So, primary carcinoma of liver was restrained.
