Object: Rheumatoid arthritis (RA) is a common difficult clinical disease, which influents sufferer' s normal life quality and even can cause disability. Until now there is still no effective and little-toxic side effect medicine in treating RA. RA belongs to "Bi-syndrome" of traditional Chinese medicine (TCM). TCM has unique understanding about etiological factor and treatments of RA. Based on the principle of strengthening vital Qi and eliminating pathogen in the Nei jing, this topic taked Bi Tong Ding capsule (BTDC) that included some valid Chinese herbal medicine certificated by clinic and pharmacology. And through modern Chinese traditional medicine pharmacology experiment, it took AA big rat model built by the complete Freund' s adjuvant to imitate the pathology variety of the RA of the mankind and observe BTDC's influence to immunology, pathology , inflammation and ache. To explore BTDC s function mechanism on treating RA and look for medicine, which is no poisonous and can be used for a long time on clinic, this topic laid the good foundation to the further development of BTDC. Method: According to the typical clinical symptom and data of RA lately, it results easily in sufferer's immunity out of tune, the joint ache or break, and it' s pathology characteristic is the chronic synovitis and the hyperplastic capillary vessel. So this project used mice to make 11 experiments from five aspects to value the curative effect and function mechanisms of BTDC on immunology, organization pathology, anti-inflammation, analgesia and acute toxic effect completely. Mice were divided into 5 groups randomly as follows: normal group, medicine control group, BTDC low group, BTDC middle group, and BTDC high group. The experiments were designed to observe following subjects: 1.The experiments on immunology ,include: the influence on the big rat serum IL-1β and TNF- a and original outbreak change of the AA big rat. 2. The organization pathology research: The influence on the big rat' s joint symposium. 3.The experiments on anti-inflammation, include the inhibition to mouse' s auricle swelling, the influence on mouse' s abdominal capillary permeability, arthritis induced with egg white and formaldehyde, and flesh turgescence induced with tampon. The experiment on analgesia. The influence on mouse' s pain threshold value in distorting body method and heat plate method. 5. The experiment on the acute toxic observing. Observing the influence of the BTDC on mouse' s physiological function. Result:1. The evaluation and the analysis of the animals model: According to the model mice' s change of the external appearance, weight, movable degree, the pathology of the joint and the whole body in the experiments,we can get such conclusion: The animal model was very successful. The animal model is generally accepted in world. It can imitate pathologic change of rheumatism arthritis and has the actual experiment value.2. The comparison and the analysis of the level of the IL-1β , TNF-α in each big rat group: Through the experiment we can discover, in the covariance, that the every treatment group can lower remarkably the level of the IL-1β , TNF-α in big rat, and compared with the model group, P<0. 05. Compared in the BTDC low, middle, high group and Zheng Qing Feng Tong Nin (ZQFTN) group, there were no obvious differences. All the above showd that the medicine of the treatment group had the repress effect to the immunity reaction of the animal model and there were the same functions between the BTDC and ZQFTN.3. The comparison and the analysis of the synovium pathologic slice of each group of the big rat' s joint in the experiment: we can get such conclusion through the observation that the treatment group had improvement function to the pathology change of the joint synovium of the model animal group.4. The experiments of the anti-inflammation, analgesia and acute toxic effect shown that BTDC could inhibit mouse' s auricle swelling by xylem, and drop the degree of inflammation bleeding of mouse' s capillary vessel, enhanced mouse...
