Background and objective Modern experiment research indicate that Codonopsis can ameliorate cellularenergy metabolism when cells are deprived of oxygen and blood. Codonopsis canimprove distinctly economy ability against deprivation of oxygen, inhibit freeradical's damnification after ischemia。Morris water maze that is designed byEnglish psychologist- Morris in the 1980s is applied to research mechanism oflearning and memory of brain.In the recent years, Morris water maze had been theimportant tool of examining experimental animals' learning and memory levels.Codonopsis can save and restore hign nerval function of the brain. Free radicalmean atom and molecule that have unpaired electron on electrical orbit. Moreexperiments indicate that Free radical can destroy proteins in the cell membrane,that result in lipid peroxidation injury, mitochondrial functional obstruction, celldissolution and tissue edema. In the order state, free radical that produce in thebody can be eliminated by superoxide dismutase(SOD)and glutathione peroxidase(GSH-Px)。Codonopsis have protective effect on ischemic tissue against freeradical injury.But it isn't known that Codonopsis abstracts(TSC and LRT-1)havesimilar effect. We establish the model by repeating transient bilateral carotidartery occlusion followed by intraperitoneal injection of 2.5mg/kg SNP. And weinject TSC,LRT-1 and PNS via rat's trail vein. We observe the effects of threeinjections on learning and memory ability of brain ischemia and reperfusion injurymodel of rat, and explore the mechamism of effects.methodFIRST EXPERIMENT:THE INFLUENCE OF CODONOPSIS TO LEARNING ANDMEMORY ABILITY OF ISCHEMIC REPERFUSION MODEL RATS To establish the model by repeating transient bilateral carotid arteryocclusion followed by intraperitoneal injection of 2.5mg/kg SNP. (2)40 Wistar ratswere devided into 5 groups randomly:①sham operation group;②operarion group;③PNS;④TSC;⑤LRT-1group. And to observe the learning and memory ability ofrats by Morris water maze, then observe pathological change through H.E stainmeans.SECOND EXPERIMENT:THE INFLUENCE OF CODONOPSIS TO SOD AND MDAOF ISCHEMIC REPERFUSION INJURY MODEL RATS To establish the model by repeating transient bilateral carotid arteryocclusion followed by intraperitoneal injection of 2.5mg/kg SNP. (2)40 Wistar ratswere devided into 5 groups randomly:①sham operation group;②operarion group;③ PNS; ④ TSC; ⑤ LRT-1group.And to observe the content of superoxidedismutase(SOD) and Malondiadehyde (MDA) by actinochemistry way.result1 In Morris water maze experiment, the swimming time until the rats arrivedplatform was prolonged significantly in the model rats as compared with the shamoperation rats(P<0.01).Compared with the model rats,the swimming time wasshorted evidently in the PNS and LRT-1 rats(P<0.05). In the space exploreexperiment, the times of across platform in 120 seconds in the model rats(4.38±2.39)were short than the sham operation rats(5.50±2.56), TSC(5.13±3.04),LRT-1rats(4.88±2.42) and PNS rats(4.63±2.07). But all is no statisticalmeaning(P>0.05).2 Compared with the sham operation rats(7.8260±1.1171), in the model rats(4.5545±1.0348)the SOD of brain tissue was decreased significantly(P<0.01),The LRT-1(6.5088±1.3655) and PNS(6.6584±0.9169)can improve SOD'scontent; Compared with the sham operation rats(4.7379±0.8051), in the modelrats ( 6.4625 ± 0.9641 ) the MDA's content of brain tissue was increasedsignificantly(P<0.01), The LRT-1(5.4476±0.6188) and PNS(5.1771±0.9504)can decrease MDA's content; But TSC can't do it.conclusion1 The brain ischemia and reperfusion model by repeating transient bilateral carotidartery occlusion followed by intraperitoneal injection of 2.5mg/kg SNP can resultin rat's learning and memory ability depression.2 The learning and memory ability depression of model rats is reversible partly.3 LRT-1 and PNS has protective effects on learning and memory ability of brainischemia and reperfusion injury model of rat.4 TSC hasn't protective effects on learning and memory ability of brain isch...
