Objective To investigate the renal protection of CCV and its mechanism inkidney ischemia/reperfusion. to provide the important and scientific basis for thenew treatment of acute renal failure.Methods 32 rats were divided into four groups. 0.1% CCV was separatelyinfused into abdominal cavity at 0.5h,24h before reperfusion in groupⅠandⅡ.Group Ⅲsufferded kidney ischemia/reperfusion as pathological control. GroupⅣ was sham operation group,used as normal control. Renal function wasmeasured by Bun and Scr. C3 was observed at 0,0.5,2,24h after reperfusion. Thekidney samples were examined by HE stain under mirror microscopy. Apoptosiswas detected by Terminal Deoxynucleotidyl transferase Mediated dUTP--BiotinIn Situ Nick –End Labeling(TUNEL).Results Significant histological damage,apoptosis of tubular cell and impairedrenal function were found in groupⅠand Ⅲ,but the change between two groupswas not significant(P<0.05). Compared with them, the above indexes wererelieved obviously in number or degree in group Ⅱ(P<0.05). it occurred ingroupⅡ that C3 was reduced to a very low level at 0h after reperfusion (P<0.05compared with the 0h level of other groups )and hadn't decreased more later.The decrease of serum C3 was not significant at 0h after reperfusion in groupⅠand Ⅲ, but there was a remarkabe decrease at 2h (P<0.05, compared with 0hseparately ).Conclusion Ischemia/Reperfusion can lead to significant histological damageand renal dysfunction. Complemental activating plays an important role in thisprocess. Pretreating the rats with CCV 12h before reperfusion can release C3notely and prevent the kidney from ischemia/reperfusion injury efficientlyinduced by complemental activation.
