| Objective: Establishment of multidrug-resistant cell line of cancer in vitro is a very important method of studying its chemothepeutic resistance. We established a Cisplatin(CDDP)-resistant human gastric cancer cell line BGC-823/CDDP and study its biological characteristics. Methods: BGC-823/CDDP was developed by a long-term continuous expousure of BGC-823 cells to CDDP in stepwise increasment of concentration.The morphological feature ,ultra structure and growth characteristics were examined. The cell cycle distribution of the two cell lines was determined by Flow Cytometry (FCM).The drug sensitivity was measured by Methythiazolyl tetrazolium(MTT) assay. Results: The multidrug-resistant cell line BGC-823/CDDP had alterd its biological properties,including changed morphology under lightmicroscopy and electronmicroscopy,faintly lower fraction of cell cycle distribution in S phase and a lower growth rate than the parental BGC-823 cells . The population double time was 4.3h longer than the parental cell line BGC-823.The resistance index of BGC-823/CDDP cells to CDDP was 11.43 and it also showed cross-resistance to Mitomycin C (MMC), 5-fluorouracil(5-Fu) and Adriamycin(ADM),the resistance index was 4.81,4.94 and 10.45,respectively. Conclusion : A Cisplatin-resistant gastric cancer cell line BGC-823/CDDP is established .It can be used for further experiments. |
