Purpose Intended to judge the feasibility and the security ofvas deferens occluding by using HIFU machine possessingindependent intellectual property right . Materials and Methods The dog's vas deferens locatingscrotum were clipped under the skin by claw through the finger ofoperator fixed vas deferens. The vas deferens in target zone wasradiated transcutaneously by transducer linking a high intensityfocused ultrasound machine designed by ultrasound institute ofchongqing university of medical science. We ablated 20 vas deferensof homing dog using a range of power and time parameters. 2 caseswere obtained immediately for pathological analysis. 2 cases wereobtained for pathological analysis at 3rd. After 2 weeks, ablation 16vas deferens, epididymis and testis was surgically harvested en bloc.Retrograde vasography and pouring water were performed to assessvas deferens occlusion; semen analysis was performed to assesswhether or not sperm existing, followed by pathological analysis. Results Post-operation, coagulation necrosis was found instantlyin histology of dissepiment ; acute inflammatory reaction wasmanifest in vas deferens after 3 days; after 2 weeks, fibrosis andcicatrization were noted in all ablated specimens, which 9 cases weredeep damage, 1 cases were moderate damage and 4 cases were milddamage. (0-1/3 section of tube injured is mild damage; 1/3-2/3 sectionof tube injured is moderate damage; 2/3-1 section of tube injured isdeep damage). There were 10 cases tube in entire occluding, 4 cases inpart occluding and no case in patent. Through semen analysis, 4 caseshad no sperm and 1 case had sperms in 5 canines whose bilateral vasdeferens occluded. In all specimens, burning of the skin was noticed,which 4 cases were mild, 2 cases were moderate and no case was deep.Bilateral sham procedures in a control dog resulted in patent duct andno associated skin burns. Conclusions The feasibility and security of occluding the vasdeferens were demonstrated through noninvasive transcutaneous highintensity focused ultrasound with ablation powers at some tested range,that are 4.8wt/cm2×10s and 5.0wt/cm2×8s. Refinement of thistechnology might provide a rapid noninvasive alternative toconventional vasectomy. Purpose Through observing the change of vas deferens andtestis, detecting proliferation and apoptosis among spermatogeniccells of dogs experiencing vasectomy at early stage; to find out thecause of lower pregnant rate in vasovasostomy, to obtain the data ofbalance mechanism in sperm generating and apoptosis. Materials and Methods 24 male adult homing dogs wereequally divided into 4 groups (6 dogs in each group) provided byanimal center of chongqing university of medical science. In eachgroup, 3 dogs were undergone vasectomy and another 3 dogs werenot undergone vasectomy. Vasectomies were done on the basis of theclinical standard for man. The duct at scrotum was resection about 1centimeter and two broken ends were ligated. The vas deferens andtestis of dogs in every group were obtained at 2, 4, 6, 8 weeks aftervasectomy. The vas deferens of dogs in control groups were onlyexposed not experienced vasectomy. All tissues were analyzed bylight and electronic microscopy. Immunohistochemical method wasused to detect the expression of Bcl-2, Bax and PCNA protein in thetestis of canines performed vasectomies and sham-operated. TUNEL was used to detect the location of apoptosis cell in testis. Results The duct of orienting testis side display duct expanding,the layer of epithelia decreasing, microvillus and vesicle of columnarepithelium cell reducing by light microscope observed; columnarepithelium cell of duct show microvillus shorting, outline of nucleusbecoming abnormal and dispersing under electronic microscope.Change of the duct orienting abdominal side was found under lightmicroscope. The number of microvillus and top bubble of columnarepithelium cell duct decreased under electron microscope.
