Objectives:Through the detection of the expression level of interleukins-1β(IL-1β), matrix metalloproteinase 9(MMP-9) and nuclear factor kappa B(NF-κB) in Synovium and the detection of the concentration of IL-6 and TNF-αin serum of collagen induced arthritis (CIA) Wister rat, to investigate the relationship between IL-1β,MMP-9,NF-κB and arthritis index(AI) in the development of arthritis,and to understand the pathology of synoviocyte.This may provide an experimented proof for the diagnosis of clinical rheumatical arthritis.Methods: Thirty male Wistar rats were randomly divided into two groups,namely the model controlled group and the normal controlled group,three subgroups were randomly divided in model group and controlled group separately.And the model group rats were immunized with emuisified bovine typeⅡcollagen in complete Freund'adjuvant by introdemal injection to induce CIA on day1,day14 and day 28,while the controlled group were injected with saline at the same time.The model and the controlrats were killed at different time.We detected the expression and activation level of IL-1β,MMP-9 and NF-κB in synovial tissue by immunohistochemistry and the concentration of IL-6 and TNF-αin the serum by ELISA.The we analyzed the relationship between AI and the expression of IL-1β,MMP-9 and NF-κB in the synovium and IL-6 and TNF-αin the serumResults: 1.CIA was established two weeks after the collagen was injected,and we can see the apparente swelling in the ankles of CIA rats.The volume of the arthritis and AI were measured 2,4,6 weeks after the models were established.Compared with the control group.AI and the quantity of the volume of the model group were higher with significant difference (P<0.001)2.1.The AI of the model group was 6.80 ,and The AI of the control group was 0.00 .there was significant difference between the model group and the contral group in AI.The volume of group in model were significantly increasing than the control. 2.Compared with the control group,IL-1βand NF-κB in nuclear of synoviocyte were increased in model group with significant difference (p<0.005);MMP-9 in synovium of model group had no significant difference compare with control group (p>0.05); 3.The concentration of IL-6 and TNF-αin the serum of model group was increased compare with control group with significant difference(P<0.05). 4. The AI in rat experimental arthritis model show a positive correlation with IL-1β(r=0.604,P<0.05),NF-κB p 65(r=0.593,P<0.05),IL-6(r=0.698,P<0.05),and TNF-α(r=0.604,P<0.05); The AI in rat experimental arthritis model don't show correlation with MMP-9...
