Both polyssacharide and selenium have the function of prevention and treatment of many diseases. The researches on the activities of polyssacharide and selenium have been performed deeply.Selenium containing polyssacharide in Ganodema Lucidum (SeGLP) is a new organic compound, it has been found has many physiological activities, such as resist of heavy mantal poisoning and active oxygen injury, anti mutagenesis and anti-tumor.The researches on selenium containing polysaccharide in garlic, Ruoye and spirulina have been peformed deeply.But the researches on extract of selenium containing polysaccharide from Ganodema Lucidum's mycelium and anti-tumor activity still stay on the primary stage in the world.Our results showed fistly that SeGLP-2B,SeGLP-3B had the fuction of inducing K562 cells apoptosis.K562 cells were incubated with SeGLP-2B,SeGLP-3B in vitro at different concentrations for 72 hours respectively. Effects on K562 cells proliferation of SEGLP-2B ,SeGLP-3B were measured by MTT assay , the morphology of K562 was observed using light microscopy and transmission electron microscopy, DNA-fragment assay was analyzed by the agarose gel electrophoresis, and the DNA contents and apoptosis cells ratro were detected by Flow cytometer.The results indicated that :SeGLP-2B, SeGLP-3B could significantly inhibit proliferation of K562 cells in dose dependent manner, the IC50 of SeGLP-2B and SeGLP-3B are 0.031g·L-1 å'Œ 0.057g·L-1.Cell crenation , condensation and margination of nuclear chromatin by cytoplasimic hypervacuolization could be seen by morphological observation.Typical DNA ladder could be seen by agar-gel electrophoresis. The results of flow cytometer showed that apoptotic peak exsisted and the sub-G0/G1 stage arrest in K562 cells treated with SeGLP-2B and SeGLP-3B.The molecular mechanisms of apoptosis of K562 cells induced by SeGLP-2B,SeGLP-3B also were studied preliminarily. Caspase-3,caspase-8,caspase-9 activities were determined by Colorimetric Assay Kit.Inhibition of caspase-inhibitors on apoptosis induced by SeGLP-2B,SeGLP-3B were detected by MTT assay, and the expression of procaspase-3, procaspase-8, procaspase-9 were detected by Western blot.In...
