Both polyssacharide and selenium play an role in prevention and treatment of many diseases. The researches on the activities of polyssacharide and selenium have been performed deeply, but it still stays on the primary stage in the world to combine the polyssacharide and selenium into selenium containing polyssacharide organically. Selenium containing polyssacharide in Ganoderma Lucidum (SeGLP) is a kind of half natural and half synthesized polyssacharide of Fungi enriched selenium, whose structurediffers from other ordinary polyssacharides——it contains a special bond betweenselenium atom and oxygen atom. SeGLP has many physiological functions, including that it can resist heavy mantal poisoning and active oxygen injury, can influence cell cycle and inhibit the synthesis of DNA of the cancer cell, and induce the apoptosis process of tumor cell.Selenium containing mycelium of Ganoderma Lucidum could be obtained by biotransformation. The mycelium was exstracted with hot water and alkali, and raw selenium containing polyssacharide (SeGLP) was obtained. Then the raw SeGLP was handled with protein and pigment removal and precipitation, and was purified and isolated by DEAE-cellulose column chromatography, the sub-fractions of raw SeGLP were obtained. Then the rough extracts passed through the DEAE-cellulose 52 and HPLC, 10 SeGLPs were obtained. The results of anticancer test in vitro showed that SeGLP-2B-1, SeGLP-2B-2 and SeGLP-2B-3 can inhibit the growth of K562 cells, and the inhibition rate of SeGLP-2B-l, about 61.58%, was highest in all samples.This paper tried to explain bioactivities of SeGLP, through contrasting chemical structure distinction between purified GLPs and SeGLPs. The experimental meterials are SeGLP-2B-1, SeGLP-2B-2, GLP-2B-1, GLP-2B-2. The infrared spectrum, ultraviolet spectrum, gas chromatogram, mass spectrum and nuclear magnetic resonance have been used in analysis of structural difference in GLPs and SeGLPs .The molecular weight was confirmed by the High Performance Liquid Chromatogram(HPLC) . MS of SeGLLP-2B-l was 1.06 × 106D ,and MS of SeGLLP-2B-2 was 1.42×104D.The atom Se of SeGLP maybe exist on chemical band Se=O or C—O—SeO3 by infrared spectrum.
