Objective:(1) To investigate the relationship between TGF-β1-induced epithelial-to- myofibroblast transition (EMT) of human kidney tubular cells (HKC) and its cellular smad mediated signalling pathway.(2) To investigate the effect of 1,25-(OH)2VitD3 on the production of fibronectin induced by TGF-β1 in renal interstitial fibroblast.(3) To investigate the effect of 1,25-(OH)2VitD3 on obstructive nephropathy.Methods:(1) Using the epithelial marker E-cadherin expression and de novo expression a-smooth muscle actin(a-SMA), the key marker of mysfibroblast, as the parameters for EMT. The TGF-p1 initiating Smad signaling pathway was tested by means of cell culture, western blot analysis and gene transfer etc. The Smad 7 expression plasmid (pSmad 7) that contains full-length human Smad 7 cDNA was transfered to HKC, to study the effect of overexpression of Smad 7 on the action of TGF-p1 on HKC.(2) Using the de novo expression of a-smooth muscle actin(α-SMA), the key marker of myofibroblast, as the parameter for renal interstitial...
