PartⅠThe expression of Prohibitin in renal tissues of rats with unilateral ureteral obstructionOBJECTIVE To investigate the expression and effect of Prohibitin (PHB) in renal tissues of rats with unilateral ureteral obstruction (UUO).METHODS Forty-eight Wistar male rats (6-weeks-old) were randomly assigned into 2 groups: sham-operated group (n=24) and model group (n=24). The left ureter of rats in mode group were dissociated to be obstructed. The rats in sham-operated group were dissociated left ureter only, and not to be obstructed. Six rats of both two group were killed on 7, 14, 21, 28 days after surgery respectively. Histological changes of renal tubular interstitium were observed by HE and Masson staining, and the index of renal interstial fibrosis (RIF) was calculated. The expressions of protein and mRNA of PHB and transforming growth factor-β1(TGF-β1) were detected by immunohistochemistry and realtime reverse transcription polymerase chain reaction (RT-PCR). The expression of protein of collagen-Ⅳ(Col-Ⅳ) and fibronectin (FN) were detected by immunohistochemistry. RESULTS (1) Compared to sham-operated group, at the same point of time:Histological changes showed that there was more collagen deposition, fibroblast proliferation and diffused lymphoeytein filtration in the renal interstitium, the index of RIF was increased remarkably in model group (P<0.01), and it was increased accompanying with the degree of obstruction. The expressions of protein and mRNA of PHB were decreased significantly in model group (P <0.01), and it was decreased accompanying with the degree of obstruction. The expressions of protein and mRNA of TGF-β1 were increased remarkably(P<0.01), and it was increased accompanying with the degree of obstruction. The expressions of protein of Col-Ⅳand FN were increased significantly (P<0.01), and it was increased accompanying with the degree of obstruction. (2) Correlation analysis showed that: the expression of protein of PHB in model group was negtive correlation with the index of RIF(r= -0.825, P<0.01), The expression of protein of TGF-β1, Col-Ⅳand FN were positive correlations with the index of RIF (r =0.995, 0.977, 0.969, P <0.01 respectively), and the expression of protein of PHB was negative correlation with the expresssion of TGF-β1, Col-Ⅳ, FN (r= -0.786, -0.948, -0.953, P<0.01 respectively).CONCLUSION The expressions of protein and mRNA of PHB were decreased significantly in renal tissues of rats with UUO, and they were decreased remarkably accompanying with the degree of obstruction. PHB maybe involved in the progression of RIF, and it's mechanism was made the expression of TGF-β1. PartⅡThe effect of all trans retinoic acid on the expression of Prohibitin in renal tissues of rats with unilateral ureteral obstructionOBJECTIVE To explore the effect of all trans retinoic acid (ATRA) on the expression of PHB in renal tissues of rats with UUO.METHODS Seventy-two Wistar male rats (6-weeks-old) were randomly divided into 3 groups: sham-operated group (n=24), model group (n=24), ATRA group (n=24). the first day before operation, the rats in ATRA group were administrated by intregastric at the dose of ATRA (15mg/kg.d) daily. Sham-operated group and model group were administrated by saline in the same volume. Six rats of the three groups were killed on 7, 14, 21, 28 days after surgery respectively. Histological changes of renal tubular interstitium were observed by HE and Masson staining, and the index of RIF was calculated. The expressions of protein and mRNA of PHB and TGF-β1were detected by immunohistochemistry and realtime RT-PCR. The expression of protein of Col-Ⅳand FN were detected by immunohistochemistry.RESULTS Compared to model group, at the same points of time: in ATRA group, the severity of interstitial fibrosis was alleviated significantly, the index of RIF was decreased remarkably (P<0.01). The expressions of protein and mRNA of renal PHB were increased remarkably (P<0.01). The expressions of protein and mRNA of TGF-β1 were decreased significantly than those in model group (P<0.01). The expressions of proteins of renal Col-IV and FN were decreased remarkly (P<0.01).CONCLUSION ATRA might increase the expressions of protein and mRNA of PHB, decrease the expressions of protein and mRNA of TGF-β1, such as a mechanism to inhibited the progression of RIF in rats with UUO.
