[Objective] The aim of the paper was to study influence of monoconjugated bilirubin(MCB) to bile nuclestion time(NT) and nucleation active(NA). To explore the mechanism of cholelith formation .[Methods] With high-performance liquid chromatography (HPLC), after being separated, paritied, prepared,the MCB was distilled from the bile.The bile was added after MCB of different concentration was put into the bile,then observe the change of NT and NA with 32 cholocystolithiasis and non- cholocystolithiasis .In different temperature,PH,the solubility of MCB was compared with the others. The cholelith was separated according to the content of cholessterin.[Results] In the bile of the cholesterol calculus the NT is 4.71 ±0.98 days,3.87± 0.74 days,2.53±0.76 days. In bilirubin calculus is 8.34±1.63 days, 7.02±1.42 days, 6.20±1.71 days. In acululus bile is 7.95 ± 1.56 days, 5.98±1.33 days, 5.07 ± 1 27days.There is significant differrence between experimental and comparative groups(p<0.05). In PH 7.9, 37 centigrade,the solubility of MCB is 559.04±4.30uM, one to six as much as the diconjugated bilirubin 's(DCB), forty five times more than unconjugated bilirubin's (UCB), In PH 7.9 it's in the summit of the solubility culve. in PH 4.5,It's zero.[Conclution] HPLC as a kind of advanced technology has been widely ussd in puriging orgainizm including MCB . When dose was doubled ,NA was stron ger than glucoprotein .MCB ,as a kind of ingredient of CB ,The insolubility was more significant than UCB,under the nomal biliary tract physiological condition. The experiment has proved that MCB has obvioussly concerned with NA.MCB has played an important role in formation of cholelith. In bile nuclearation the MCB is a significant ccomponent.
