Objective : To investigate the influence of mifepristone on the expressions of COX-2 and CDK4 in the xenograft of endometrial carcinoma HHUA.Methods: The twenty nude mice with xenografts of endometrial carcinoma HHUA were established and divided randomly into two groups, the mifepristone group and control group. The nude mice of mifepristone group were feeded with the dose of mifeprisotne 50mg.kg-1.d-1, and the nude mice of control group were given peanut oil for 6 weeks, respectively. The growth and volume of xenograft were measured in the pre- and post-treatment. The changes of xenograft morphology were observed by light microscopy with HE staining.The intensity and the distribution of COX-2 and CDK4 expression were analyzed by an image-pro plus 4.1.00 immunohistochemical scoring system (HSCORE).Results: After 6 weeks of treatment, the size of the xenografts of mifepristone group was 115.25 ± 10.97mm3, which was significant smaller than 313.25 ± 43.92 mm3 of control group (P<0.01). The xenograft of mifepristone group appeared significantly necrosis morphology changes. The COX-2 HSCOREs of mifeprisotne group were 78.2 ±11.3, which were lower than 205.9±26.7 of control group (P<0.01). The CDK4 HSCOREs of mifeprisotne group was 113.4 ± 18.2, which were lower than 238.7±35.9 of control group (PO.01).Conclusion: Mifepristone inhibites significantly the growth of the xenografts of endometrial carcinoma HHUA in nude mice by down-regulating the expressions of COX-2 and CDK4.
