Objective:To explore the effect of intervention with Alarelin on TRF2 expressions in endometrial cancer and subcutaneously xenografted tumor in nude mice.Methods:The HEC-1-B cells people endometrial carcinoma transplanted subcutaneously in nude mice tumor model was successfully established.And we selected 32 female bearing nude mice,which were randomly divided into 4 groups, namely:blank control group (normal saline 0.2ml), leuprolide acetate Ruilin intervention group (20 u g/kg,40 g/kg and 80 mu g/kg). The growth and development of nude mice should be observed every 7 days.We measure subcutaneous transplantation tumor of the long diameter and short diameter, and calculate the volume. After 28 days, the mice were killed, removed the transplanted tumor from the mice, and calculation tumor inhibition rate. Transplanted tumor tissue embedded in paraffin and stained with hematoxylin eosin (HE) staining, the tumor cells were observed under microscope. The expression of telomere repeat factor 2 (repeat binding factor-2 TRF2, Telomeric) in tumor tissues was detected by immunohistochemistry. SPSS 19.0 software was used to analyze the all data. The measurement data were expressed by x±s,and the intra-group comparison was tested by LSD t-test whereas group-group used variance analysis.Each group was compared with one-way ANOVA P<0.05 was considered to be of statistical significance.Results:The blank control group, low, middle and high acetic acid bright propylene Alarelin group transplanted tumor growth curve slope decreases, growth slowed. The tumor inhibition rates were:low measurement Alarelin group 10.01%, middle dose Alarelin group 19.80%, high dose Alarelin group 44.98%. There were significant differences between the two-two groups (P<0.05). After HE staining, the tumor cells in the intervention group were less than those in the control group, and the cell nuclei were stained and contracted. By detecting TRF2 in immunohistochemical, positive staining is located in thenucleus or the cytoplasm, mainly located in the cell nucleus, which showing brown-yellow granules.With the increase of the drug dose, the TRF2 was up-regulated in the tumor tissue, and the difference was statistically significant (P<0.05) in the Alarelin group compared with the control group.Conclusion:Alarelin have direct inhibitory effect on nude mice who bearing human endometrial carcinoma. The mechanism of down-regulation of the TRF2 may be through which Alarelin inhibited the activity of telomerase, damaged the vigour of tumor cells, and then achieved the purpose of inhibiting tumors.
