Objective:Through the establishment of endometrial carcinama in nude mice subcutaneous transplantation tumor model,with different doses of propylene ammonia gonadorelin group intervention,to a tumor-burdened nude mice to observe different doses of propylene ammonia gonadorelin intervention of endometrial carcinoma in nude mice subcutaneously transplanted tumor growth and the Fli-1 protein expression in transplanted tumor. Methods:Nude mice were inoculated with the human endometrial carcinoma cell line HEC-1B,that has culturaled in vitro.Building the model of human endametrial cancer xenogreft,32 female mice were randomly divided into four groups: the low-dose group(20μg/kg),the medium dose(40μg/kg),the high-dose group(80μg/kg),and the blank control group(0μg/kg)..Executing mice four weeks later, and the tumor was removed completely, after that, measuring the volume and calculating the inhibition rate, detecting the expression of Fli-1 in tumor tissue with immuno-histochemical. Results:(1)The common situation of the experimental miceNude mice’ reaction and activity is good and the weight doesn,t change obvious in the process of from selected to human endometrial carcinoma model established; In the process of tumors had gradually increased, reaction force and the activity of the nude mice gradua l decline downdrift, and the weight reduce gradually. Reaction force and the weight of eac h treatment group nude mice higher than the control group with different levels.(2) The volume of tumors after different doses of drug interventionIncrease with concentration of Alarelin,tumor growth gradually slowed, tumor growth is restrained obviously. The volume of tumors of the control group,Alarelin low, medium a nd high dose group product respectively(1812.93± 196.249 mm3),(1518.82±124.933 mm3),(1399.44±187.657 mm3),(1164.55±189.118 mm3), pairwise comparisons among groups employing the LSD-t test, statistical significance differences were found between each two groups(F= 18.604, P <0.001). The inhibitory rate of low, medium and high dose group, res pectively(14.76± 4.904%),(27.5±5.456%),(36.77±2.67%), the LSD- t test pairwise comp arisons between the experimental way, the investigators observed statistically significant(F =31.477, P < 0.001). Show that Alarelin can inhibit tu mor growth, and has the obvious dose-dependent.(3)The hematoxylin-easin staining resultsAlarelin treatment group sparse arrangement of tumor cell and slightly larger, some ce lls pyknosis, cracking,and cell apoptosis and necrosis can be seen; Control tumor cell diffu se distribution, border of neoplastic cell,s clump were clear,deep-dyed big nucleous, nucle oplasm ratio im balance, more nuclear fission can be found.(4) Immuno-histochemical resultsFli-1 protein expression in transplanted tumor reduce after alarelin intervention. Grey value analysis for each slice products found in the control group, alarelin low, medium and high dose group of transplanted tumor tissue protein the Fli-1 respectively:(118.1±4.241),(135.93±3.548),(147.51±6.755),(168.84±5.202),compared with the control group,the grey value expression level of the Fli- 1 protein in the treatment groups also increased to diffe rent extent, and the comparison between different dose groups using LSD-t test difference was statistically significant(F = 140.196, P < 0.001). Conclusions:(1) Alarelin can restained the growth of endometrial cancer xenograft,with propylene ammonia gonadorelin dose add inhibitory effect more obvious at the same time.(2) Alarelin tumor inhibitory mechanism may be related to the down-regulated expression of Fli-1 protein.
