Purpose : To observe whether it can inhibit the growth of pancreatic carcinoma cells in vitro and vivo experiments with NC-8-12(specific agonist of somatostatin receptor II) by transfecting pancreatic carcinoma cells and pancreatic carcinoma hypodermic model of naked rat with a recombinant adenovirus vector , named as Adeno-X-SSTR2, which takes SSTR2 gene.Methods: SSTR2 gene was cloned and recombined into Adeno-X-SSTR2 gene,to observe the inhibition of NC-8-12 of various dose (10-8M, 10<sup>-9M) to pancreatic carcinoma cells of transfected and untransfected SSTR2 gene by infecting MIA-PaCa-2, AsPC-1 separately in vitro ;constitute hypodermic model of naked rats with infecting and uninfecting SSTR2 gene separately, which were given NC-8-12 of various dose (50 , 100 , 200ug/kg/d) by hypodermic injection for 3 weeks, and then killed the rats 1 weeks later, to observe the change of tumor volume and weight by taking normol saline group as control group.inspecting the expression of SSTR2 in tumor tissue by immunohistochemistry technique.Results: NC-8-12 has no inhibition to pancreatic carcinoma cells with untransfecting SSTR2 gene in vitro experiment (P>0.05). NC-8-12 can inhibit tumor cells proliferation induced by insuline signifiently at the dose of 10-8M (P<0.05). NC-8-12 has no inhibition to pancreatic carcinoma cells without transfecting SSTR2 gene in hypodermic model of naked rats (P>0.05). NC-8-12 can inhibit tumor cells proliferation remarkably at the dose of 200μ g/kg/d in the cells transfected SSTR2 gene (P<0.05).
