Four Chinese Medicine Monomer On The Role Of Hepatic Stellate Cells | Posted on:2007-07-09 | Degree:Master | Type:Thesis | Country:China | Candidate:W L Zhang | Full Text:PDF | GTID:2204360185488313 | Subject:Digestive medicine | Abstract/Summary: | PDF Full Text Request | Part 1 THE ISOLATION AND CULTURE OF RAT HEPATIC STELLATE CELLSAims: To isolate and get high output and purity rat hepatic stellate cells Methods: The rat hepatic stellate cells were isolated with collagenaserecirculating perfusion and purified with density gradient centrifugation.Resultes: In this way, the output of the hepatic stellate cells amounted to5-6×107 cells per liver. The purity of the cells was above 97%. The viability of the cells was more than 92%.Conclusions: Getting high output and purity hepatic stellate cells.Part2 OBSERVING THE EFFECTIVENESS OF DANSHENSU,ASTRAGALUS,BAICALIN AND SAGIOEM ON HEPATIC STELLATE CELLSObjectives: Contrast to colchicine, to study the effectiveness of danshensu, astragalus, baicalin and sagioem on hepatic stellate cells(HSCs)Method: The rat hepatic stellate cells were isolated with collagenase recirulating perfusion and purified by density gradient centrifugation.MMT colorimetric assay was used to detect HSCs ploliferation . Flow cytometry was used to observe the cell cycles and apoptosis of HSCs.Apoptosis was also demonstrated with AO/EB fluorescence staining. Type I and type III collagen were detected by immunocytochemical staining. | Keywords/Search Tags: | Hepatic stellate cells, Cell isolation, Cell culture, Recirculating perfusion, proliferation, apoptosis | PDF Full Text Request | Related items |
