Extract. Centipede Thirty-seven Anti-research, Rheumatoid Arthritis, The Immune Mechanism

Objective: Rheumatism arthritis (RA) is one of the most familiar rheumatism. It is distributing all over the world and has a high morbidity and disability rate. The pathogeny and mechanism of RA is very compex. Abnormality of cell-mediated immunity is thought of as the key of RA. T cell is the most one between inflammatory cells in synovial joint. The immune damage which is brought by excessive activated T cell aggravates inflammatory reaction and synovial hyperplasia. Costimulatory molecules,cell apoptosis molecules and cytokines play a great important role in occuring and continuing of inflammatory reaction in RA.Wugong Sanqi is the rhizome of Goosepalm Windf lower which belongs to Trib. Anemoneae.It is used as the treatment for rheumatism pain, expelling wind-evil and removing wetness-evil. Institute of traditional Chinese medicine in our college has got extracts from Wugong Sanqi through distilling and separating. The extract has been proved to have a notable effects on RA through plenty of experimental researches in laboratory and authorized for clinical experiments as the fifth kind of new medicine. On the base of these researches, we tried to study what roles costimulatory molecules,cell apoptosis molecules and cytokines play in the course of Wugong Sanqi extract against RA in this experiment, to discuss the immune regulative mechanism of Wugong Sanqi extract against RA, to provide experimental evidence for its application in clinic on a large scale earlier.Methods: Take collagen II -induced arthritis (CIA) mouse as animal model,divid mouse into normal group,model group, tripterygium wilfordii group and Wugong Sanqi extract groups including the high dose group, the medium dose group, the low dose group. These groups were orally adminstered for 36 days. Make use of FCM, ELISA to study immune therapeutic mechanism of Wugong Sanqi extract to rheumatoid arthritis from different points of view (such as pathology, immunology, molecular biology,etc) and various administrative levels (body,tissue,molecule). Result:1 Typical diagnostic of RA arose when induced inflammation 14-16 d on CIA mouse.Pathological changes became worsen day after day and arthrocele achieved its peak 36 d. There was obvious swelling on paws and tail. Mouse were inappetent, becoming thin and their coats losing luster gradually in course of pathological changes.2 Made the grade of joints for CIA mouse. Wugong Sanqi extract groups and tripterygium wilfordii group had a obvious fall compared with model group.The low dose group was the best of all (P<0. 001).3 Pathological histology in synovial joint was observed by optical microscope.
