Objective To investigate the relationship between the proteinum expression of Mdm2, PARP,ToPoⅡ and ToPoⅡ gene mRNA in laryngeal carcinoma, and their relationship with clinic opathological feature. Methods Expression of Mdm2, PARP and ToPoⅡ protein was measured with immunohistochemistry method in 50 cases of laryngeal squamous cell carcinoma and 10 cases of normal laryngeal mucosa, and the expression of ToPoⅡ mRNA was detected by insitu hybridization in conesponding samples. Results (1)The expression of Mdm2 positive was 86% (43/50) ,20% (2/10)in laryngeal carcinoma and normal laryngeal mucosa. Mdm2 expression level was correlated with clinical stages and lymphnode metastasis,and had no correlation with sex ,differentiation level, clinical stages and T stages. (2)The expression of PARP positive was 76% (38/50),30% (3/10) in laryngeal carcinoma and normal laryngeal mucosa. PARP expression level was correlated with differentiation level , and had no correlation with sex,clinical stages and lymphnode metastasis,clinical classification. (3)The positive ratio expression of ToPoⅡ protein and gene mRNA in laryngeal carcinoma was 70% (35/ 50)and 64% (32/50), and that in normal laryngeal mucosa was 10% (1/10) and 20% (2/ 10). ToPoⅡ expression level was correlated with clinical stages, lymphnode metastasis and differentiation level. ToPoⅡ related intimating with clinic chems. And had no relation with sex,clinical classification and T stages. The expression of ToPoⅡ gene mRNA and protein had consistency(P<0. 05). (4)The three agent dependablity:The study showing Mdm2 and ToPo Ⅱ had negative correlation(r= - 0.946), PARP,Mdm2 and ToPoⅡ had no obviously dependablity. Conclusions This experiment suggested that Mdm2, PARP and ToPoⅡ may play certain roles in the oncogengsis and progession of laryngeal carcinoma. Detection of these gene products had positive offects on early diagnosis,malignancy determination prognosis and treatment of chems laryngeal carcinoma.
